Chronic spontaneous urticaria(CSU)refers to a disease in which itchy,red or pale wind masses appear in the skin and mucous membranes.The course of the disease is more than six weeks and recurrent attacks.Most patients have mild symptoms and can be healed within one year,some patients have a longer course of disease,even more than five years,and a very small number of patients may not heal for life,seriously affecting daily life.The onset of urticaria is often accompanied by the activation of skin mast cells.When human skin mast cells(MC)are stimulated by external factors,they will degranulate,which will make people feel itchy and make the skin red or pale.Colored wind group.Mast cells are stimulated to play a key role by releasing various vasoactive mediators and various proteases,chemokines and cytokines.It is understood that 30% to 50% of CSU patients have an autoimmune basis,because these patients either show autoantibodies to the high-affinity Ig E receptor Fc?RI,or show autoantibodies to Ig E(this is rare).The remaining approximately 50% of CSU patients have truly sudden conditions.This project screened the itch-related G Protein-Coupled Receptor(GPCR)and new peptides in peripheral blood leukocytes of patients with CSU,and found the expression of new peptide C17ORF67 in peripheral blood leukocytes of patients with CSU is significantly higher than that of normal people.Through further research on the degree of itching,itching time,and HE staining of the C17ORF67 gene knockout urticaria model mice,it is found that the C17ORF67 gene knockout can alleviate the itching caused by urticaria in mice,and overexpress the C17ORF67 gene Later,it will aggravate the severity of urticaria in mice.In addition,we also conducted a preliminary screening of the signal pathway of C17ORF67 in chronic spontaneous urticaria.The results showed that C17ORF67 may mediate the downstream pathogenesis by activating the JAK1/STAT3 signaling pathway.Therefore,the new peptide C17ORF67 gene may be a treatment for chronic spontaneous urticaria.A new target for patients with urticaria.
