Objectives The Complementary Acupoints",as a kind unique theory of acupoints combination,refer to the acupoints which are situation on the corresponding medial and lateral sides of the limbs or in the front and back parts of the body trunk.To observe the Mast Cells of chemotaxis and degranulation in the point local the expression of substance P?histamine and bradykinin in serum after needing the “Complementary Acupoints” Yanglingquan and Yinlingquan in rats.To discuss the mechanism and regulation of acupuncture effect after needling the “Complementary Acupoints”.To provide experimental basis for the clinical application of the “Complementary Acupoints”.Methods 40 Wistar rats were randomly and equally divided into control(group K),acupuncture the point Yanglingquan(group A),acupuncture the point Yinlingquan(group B),acupuncture the point Yanglingquan and Yinlingquan(group AB)and acupuncture the sideward of Yanglingquan and Yinlingquan(group CD),8 rats each.The control group was not treated.The intervention group were treated with electroacupuncture,once every other day,drawing materials after 2 hours.in fourth times of intervention.The changes of pain threshold were detected and compared before and after the experiment.Detection the content of substance P histamine and bradykinin in rats' serum by ELISA technique and the expression of Mast Cells function and substance P in the point local by histochemistry method after electroacupuncture.Results 1 Comparison of chemotaxis and degranulation in each groups: Compared with the group K and group CD,the positive expression of Mast Cells number and the degranulation in the local area in groups A,B,AB were increased in different degrees.Among them,the group AB was the most significant difference(P<0.01).There is no statistical significance in compared the group A with group B and the group K with group CD.2 Comparison of the the expression of substance P in each groups: Compared with the group K,the positive expression of substance P in the local area and the content of the serum in groups A,B,AB and CD were increased in different degrees.There was a remarkable difference between group K with groups A,B,AB(P<0.01).But there was no statistical significance compared with group K and CD(P>0.05).A group,B group compared with AB group,substance P expression was significantly different(P<0.01)and there was no significant difference between group A and B(P<0.01).Compared with other acupuncture groups,the substance P expression in the point local and the serum of group E was significantly different(P>0.05).3 Comparison of the the content of histamine and bradykinin in each groups: Compared with the group K,the content of the serum histamine and bradykinin in groups A ? B ? AB and CD were statistically significant changed.There was a remarkable difference of the content of histamine between group K with groups A?B?AB and CD(P<0.05),group AB was specially higher than other groups;There was a remarkable difference of the content of bradykinin between group K with groups A?B and AB(P<0.05),but there was no statistical significance compared with group K and CD(P>0.05).Conclusions After needling the “Compplementary acupoints” Yanglingquan and Yinlingquan,The significantly increased of the Mast Cells of chemotaxis and degranulation and the positive expression of substance P in the local area and the significantly increased content of histamine and bradykinin of the serum were one of the possible factors for the mechanism and regulation of acupuncture effect in “Complementary Acupoints”.At the same time that we can the Mast Cells of chemotaxis and degranulation change and the expression of SP in the local area and the content of HIS and BK as one of the reference indexes to quantify the effect of acupuncture.And providing experimental and theoretical basis for the clinical judgment index to cupuncture effect.
