| Purposes:In this experiment was to observe the effect of Acupuncture Pretreatment at Quchi and Xuehai on IL-33/ST2 expression and mast cell degranulation in urticaria rats,and to explore the effect and mechanism of Acupuncture Pretreatment at Quchi and Xuehai on urticaria rats.Material and methods:Thirty-five 6-week-old SPF male SD rats were selected by completely random method to prepare anti-ovalbumin serum.After adaptive feeding for one week,32 SD rats were randomly divided into blank control group,model control group,acupuncture treatment group and positive drug group,with 8 rats in each group,and given different intervention measures.Acupuncture group was given acupuncture intervention,acupuncture on bilateral Quchi and Xuehai acupoints,20 minutes/time,1day each time;The positive drug group was given loratadine 5ml/kg(?)d-1by gavage;Model rats were treated with saline lavage,volume,intervention,intervention for 7days in a row,and on the fifth day after using dilute of ovalbumin serum of model control group,the acupuncture treatment group and positive medicine group experimental rats back marked points back to quick build mould injection,7 day antigen attack and materials.He staining was performed on the sensitized skin tissue to observe the morphological changes of the sensitized skin tissue.The degranulation of mast cells in loose connective tissue was observed by toluidine blue staining.Serum Ig E and histamine levels were detected by ELISA.The expression of IL-33/ST2 in the sensitized area of rats was detected by immunohistochemical method.Results:1.During the pretreatment,the rats in the blank group maintained normal state,good spirit,sensitive activity and normal diet;After the intervention,the rats in the acupuncture group occasionally showed emotional agitation,the frequency was less,and the others were as usual;In the positive drug group,the activity of each rat was reduced,loose stool was occasionally seen,and others were normal.During the whole process,all rats survived without death.Within 1 hour after antigen challenge,the rats in blank control group were quiet and occasionally scratched the injection part,with less times and frequency;In the model control group,the rats were restless,scratched and rubbed their back frequently,and the activity increased significantly;Compared with the model group,the rats in the acupuncture group and the positive drug control group were obviously less scratched.2.After antigen challenge,there was no obvious blue spot on the back of rats in the blank control group,but the blue spot in the model group was obvious,and the diameter of blue spot was statistically significant compared with that in the blank control group(P<0.01);compared with the model control group,the diameter of blue spot in the acupuncture group and the positive drug control group was significantly smaller(P<0.01);there was no significant difference between the acupuncture group and the positive drug control group(P>0.05).3.Evans blue dye exudation results showed that the amount of dye exudation in blank control group was relatively less than that in model group(P<0.01);Compared with the model control group,the acupuncture group and the positive drug group decreased(P<0.01 or P<0.05),but there was no significant difference between the acupuncture group and the positive drug control group(P>0.05).4.The results of HE staining showed that:In the blank control group,the structure of epidermal cells was normal,the arrangement of collagen fibers in dermis was regular,and there was no obvious edema and inflammatory infiltration;In the model control group,the structure of epidermal layer was incomplete,the epidermal cells were disordered,the fibers in the dermis were arranged disorderly,and there were inflammatory cells infiltration and edema;In the acupuncture group and the positive drug group,the epidermal cells were arranged regularly,occasionally the epidermal structure was incomplete,the fibers in the dermis were arranged in a reticular arrangement,the gap was significantly smaller than that in the model group,and the inflammatory cells and edema were alleviated in varying degrees.5.The results of toluidine blue staining showed that the mast cells in subcutaneous connective tissue of the blank control group were relatively complete,stable and the degranulation rate was low;compared with the blank control group,the degranulation rate of mast cells in the model group was significantly increased(P<0.01);compared with the model control group,the degranulation of mast cells in acupuncture group and positive drug group was significantly reduced.The rate of depigmentation was significantly reduced(P<0.01).6.The results of ELISA showed that:Compared with the blank control group,the serum Ig E and histamine levels of the model group were significantly increased(P<0.01);Compared with the model control group,the levels of serum Ig E and histamine in the acupuncture group and positive drug group were lower than those in the model group(P<0.01 or P<0.05);There was no significant difference between the acupuncture group and the positive drug control group(P>0.05).7.The results of immunohistochemistry showed that:compared with the blank control group,the expressions of IL-33 and ST2 in the model control group were significantly increased(P<0.01 or P<0.05);compared with the model control group,the expressions of IL-33 and ST2 in the acupuncture group and the positive drug group were relatively decreased(P<0.01);there was no significant difference between the acupuncture group and the positive drug group(P>0.05).Conclusion:Acupuncture at Quchi and Xuehai points can reduce the serum total Ig E level by inhibiting the expression of IL-33/ST2,and reduce the degranulation and histamine release caused by mast cell contact in rats,so as to effectively improve the skin morphological changes and restless pruritus in the sensitized skin of urticaria rats.In order to play the purpose of treatment. |
