The Effect Of Zhuyang Tongbian Ointment On The Colonic Motility Of STC Model Mice

Purpose: To observe the therapeutic effect of Zhuyangtongbian Paste on STC model mice,and to explore the regulation mechanism of colon SP and c-kit(ICC)expression in STC model mice,and to reveal the possible therapeutic mechanism of Zhuyangtongbian Paste in the treatment of STC diseases from the perspective of cell and molecule.Material and method:After 1 week of adaptive feeding,75 mice were divided into 5groups by random number table method.Mice in each building modules group were given compound difenoxate tablets suspension 0.2ml/10 g body weight once a day for 7 consecutive days to replicate the STC mouse model.After the successful replication of the STC model,the patients in the Zhuyangtongbian Soup group were given Zhuyangtongbian Soup,Zhuyangtongbian Paste group were given Zhuyangtongbian Paste solution,and the patients in the Maren Capsule group were given Maren Capsule suspension once a day.The patients were given gastric gavage once a day.The therapeutic intervention lasted for 14 days,and the patients in the STC model control group and the blank control group were given normal saline once a day.After the final administration,the intestine carbon propulsion rate of mice in each group was determined,and the expression levels of SP and c-kit(ICC)in colon tissues of mice in each group were detected by Western-blot.SPSS17.0 statistical software was used for statistical analysis of experimental data.Results:1.24 h fecal weight:before modeling,there was no significant difference in the fecal weight of each group at 24h;after modeling,compared with the blank control group,the fecal weight of mice in the modeling group was significantly reduced at 24 h,(P<0.05).2.Carbon propulsion rate:compared with the blank control group,the carbon propulsion rate of the STC model control group was significantly lower,(P<0.01);compared with the STC model control group,the carbon propulsion rate of the Zhuyangtongbian Paste group was significantly higher,(P<0.01).3.Expression levels of SP and c-kit(ICC)in colon:3.1.Compared with the STC model control group,the SP expression was significantlydecreased in the Zhuyangtongbian Paste group,Zhuyangtongbian Soup group,and Maren Capsule group,(P<0.01);compared with the Zhuyangtongbian Soup group,the SP expression was significantly decreased in the Zhuyangtongbian Paste group,(P<0.01);SP expression was significantly lower in the Zhuyangtongbian Paste group than in the Maren Capsule group,(P<0.01).3.2.Compared with the STC model control group,the c-kit(ICC)expression was significantly increased in the Zhuyangtongbian Paste group,Zhuyangtongbian Soup group,and Maren Capsule group,(P<0.01);compared with the Zhuyangtongbian Soup group,the c-kit(ICC)expression was significantly increased in the Zhuyangtongbian Paste group,(P<0.01).c-kit(ICC)expression was significantly higher in the Zhuyangtongbian Paste group than in the Maren Capsule group,(P<0.01).Conclusion:1.Compound difenoxate tablets suspension was successfully used to replicate STC mice model by gavage.The mode of modeling is safe and reliable,with simple operation and controllable dose,and the model conforms to the basic symptoms of clinical STC patients.2.Zhuyangtongbian Paste is effective in the treatment of constipation in STC model mice.The effective mechanism of Zhuyangtongbian Paste in the treatment of STC diseases may be related to the reduction of SP expression in the colon tissue of STC model mice and the increase of ICC quantity in the colon tissue of STC model mice,and the multi-mechanism synergistic promotion of intestinal peristalsis function effect in STC model mice.3.The intervention effect of Zhuyangtongbian Paste on colon dynamic in STC model mice was more obvious,which may be related to the higher crude drug content of the paste in unit dose,and the fact that black sesame and honey are added to the paste to to assist made from cream and enhance the effect of medicine to relaxing bowel at the same time.
