Objective: To investigate the effect of Bai zhu Qi wu granules on expression of dopamine D2 receptor(Drd2)?Inhibitory neurotransmitter vasoactive intestinal peptide(VIP)and excitatory neurotransmitter substance P(SP)in the Colon Tissue of rats with slow transit constipation(STC)and to preliminarily investigate the efficacy and mechanism of Bai zhu Qi wu granules in the treatment of STC.Methods: From June to October in 2016,60 SD rats were divided into blank group(n=10),model group(n=10),positive group(n=10),low-dose group of Bai zhu Qi wu granules(n=10),Middle-dose group of Bai zhu Qi wu granules(n=10)and high-dose group of Bai zhu Qi wu granules(n=10)by random number table method,model group?positive group and low-dose group ?Middle-dose group and high-dose group of Bai zhu Qi wu granules were administrated with morphine hydrochloride injection by subcutaneous to build STC models;blank group was administrated with the same amount of 0.9% sodium chloride injection by isotonic subcutaneous injection.The general condition of the rats was recorded;before model building and 24 hours after model building,the condition of dry weight faeces and the rate of charcoal propulsion in intestinal tract were observed.After successful model building,the low-dose group ?Middle-dose group and high-dose group of Bai zhu Qi wu granules group were administrated with water solution of Bai zhu Qi wu by drench,the positive group was administrated with water solution of Itopride Hydrochloride Tablets by drench;the other two groups were administrated with the same amount of 0.9% sodium chloride injection by isotonic subcutaneous injection.The expression of Drd2 ?the content of VIP and SP in the Colon Tissue of rats were detected 14 days after drug administration.Results :The rats of the blank group had flexible motion and normal diet,drinking and defecation;the rats of the other five groups behaved sluggish in moving,with their hair dry and yellow,the amount of stool decreasing and the stool particle being smaller and harder.Before model building,the six groups were not significantly different in the number of rats having dry weight feces(p > 0.05),after model building,the model group?positive group ?the low-dose group ?Middle-dose group and high-dose group of Bai zhu Qi wu granules group were lower than blank group in the number of rats having dry weight feces and the intestinal ink advance rate(p<0.05).The model group?positive group ?the low-dose group ?Middle-dose group and high-dose group of Bai zhu Qi wu granules group were higher than blank group in the rate of expression of Drd2 in the Colon Tissue of rats(p<0.05);the positive group and the low-dose group ?Middle-dose group and high-dose group of Bai zhu Qi wu granules group were lower than model group in the rate of expression of Drd2 in the Colon Tissue of rats(p<0.05)Compared with the the rate of expression of Drd2 in the Colon Tissue of rats,There was no statistically significant difference between the low-dose group of Bai zhu Qi wu granules and positive group(p>.05).Middle-dose group and high-dose group of Bai zhu Qi wu granules group were lower than positive group in the rate of expression of Drd2 in the Colon Tissue of rats(p<0.05).After model building,the model group was higher than blank group in the content of VIP(p<0.01),the positive group and the low-dose group ?middle-dose group and high-dose group of Bai zhu Qi wu granules were lower than model group in the content of VIP(p<0.05),the low-dose group ?middle-dose group of Bai zhu Qi wu granules were higher than high-dose group of Bai zhu Qi wu granules in the content of VIP(p<0.05).After model building,the model group was lower than blank group in the content of SP(p<0.01),the positive group and the low-dose group ?middle-dose group and high-dose group of Bai zhu Qi wu granules were higher than model group in the content of SP(p<0.01),the positive group and the low-dose group ?middle-dose group of Bai zhu Qi wu granules were lower than high-dose group of Bai zhu Qi wu granules in the content of SP(p<0.01)?Conclusion: Bai zhu Qi wu granules are effective in the treatment of STC and the mechanism may be related with the reduction of expression of Drd2 in the colon tissue of rats,the reduction of VIP and the improvment of SP.
