Objective: Polygonum multiflorum is widely distributed,rich in chemical substances,high medicinal value,shortage of wild resources,and the quality of Polygonum multiflorum is quite different.There are many kinds of medicines in clinic,so the quality control and quality evaluation of Polygonum multiflorum is particularly important.By investigating and identifying the resources and varieties of wild Paris polyphylla in Northwest Hubei,routine content determination and saponin extraction determination of Paris polyphylla,a stable and reliable method of high performance liquid chromatography fingerprint of Paris polyphylla was established.Similarity evaluation,cluster analysis and principal component analysis were used to compare and evaluate the quality of Paris polyphylla from different habitats,so as to enhance the feasibility of quality control of Paris polyphylla.It can provide a basis for the selection and clinical application of raw materials of traditional Chinese medicine,provide reference for the artificial standardized planting of Zhonglou,and solve the current situation of insufficient wild resources of medicinal materials.Method:(1)Resource investigation and variety identification: including resource investigation,sample collection,identification and classification,morphological difference comparison between different species and genera,identification of different subgenera and DNA barcode identification;(2)Content determination: Referring to the method described in Appendix of Chinese Pharmacopoeia 2015,routine inspection items(water,ash,acid insoluble ash)of Paris polyphylla were determined.Orthogonal test was used to optimize the extraction and determination method of saponins from Rhizoma Paris,and the content of various saponins in Rhizoma Paris was determined by HPLC to establish objective indexes for quality control of Rhizoma Paris.(3)Quality control and quality evaluation: Establishment of high performance liquid chromatography fingerprint of Rhizoma Paris,and comparative evaluation of quality of Rhizoma Paris from different producing areas was carried out by similarity evaluation,cluster analysis and principal component analysis.The purpose is to reflect the internal chemical information of the heavy building and evaluate the quality of the heavy building in Northwest Hubei.Result:(1)27 wild Paris polyphylla germplasm resources in Northwest Hubei were collected and identified as 7 lateral membranous subgenera.Through observation and comparison,it was found that the morphological differences of Paris polyphylla of different species were obvious,and the subgenera could be identified according to the mode of ovule implantation and seed difference on the transverse section of ovary of different Paris polyphylla.The secondary structure of ITS2 contained one main ring and four helical regions,with different numbers on each helix.Different stem-ring structures can be clearly distinguished by the secondary structure of ITS2;(2)Routine inspection items(moisture,ash,acid-insoluble ash)of 29 batches of wild Paris specimens collected all meet the requirements of pharmacopoeia;the best method for the determination of Paris is to elute it with 0.05% H3PO4-acetonitrile as mobile phase gradient at 203 nm wavelength for 120 minutes,and Paris repens is a new plant.The optimum extraction method of saponins was ethanol water bath reflux twice(1.5 h,1 h);12 batches of wild Paris polyphylla were established by HPLC fingerprints;(3)29 batches of Paris polyphylla samples were analyzed accurately.The results showed that only 8 batches of Paris polyphylla samples met the requirements of pharmacopoeia,the qualified rate was only 28%,and the quality was poor,but the Majiahe Longwangzhai Paris polyphylla in Maojia area was poor.The quality is good;12 batches of wild Paris polyphylla samples were collected by high performance liquid chromatography fingerprint method to investigate the overall information of chemical components.The results show that there are 12 common peaks in the fingerprint of Paris polyphylla.The similarity analysis by similarity software shows that the similarity results are between 0.627 and 0.908.Conclusion:(1)The natural conditions in Northwest Hubei are superior,the resources of heavy buildings are abundant,and the flower of seven leaves is abundant.The unique species of heavy buildings in Zhuxi,Hubei,was identified and named by Li Heng,a scholar.The varieties of heavy buildings with bright leaves and ball Medicine found in Fulongshan base were included in "Heavy Flowers".Through observation and comparison,it is found that the morphological differences of different species of heavy buildings are obvious,which can be based on.These differences are more convenient and rapid for identification of Paris;effective identification of Paris subgenus can be carried out according to the different ways of ovule ovary implantation and seed differences on cross section of Paris ovary;ITS2 as DNA barcode sequence can effectively distinguish Paris plants;genetic distance analysis can be carried out by using ITS2 intraspecific identification research,and it can be used for germplasm species in the process of standardized planting of Paris.(2)Compared with the methods recorded in the Pharmacopoeia,the established methods have similar results,but improve the analysis efficiency,reflect more comprehensive information of Paris polyphylla,and can objectively and effectively evaluate the quality of Paris polyphylla from different origins,which can be used as a favorable means for quality evaluation of Paris polyphylla.The content of Paris saponins in different varieties from different origins in Northwest Hubei is different.The quality of Paris polyphylla from more producing areas could not meet the requirements of pharmacopoeia.All samples from the same producing area had higher content and better quality,which could be used as standard planting provenances.The newly discovered Paris polyphylla polyphylla had high research value and could be considered as a new drug source for popularization and planting;(3)The similarity of 12 batches of medicinal materials was 0.627-0.908,which could be 0.800-1.000 were grouped into one group and less than 0.7 were grouped into one group.Cluster analysis showed that 12 batches of Rhizoma Paris could be grouped into two groups,in which the relationship between Rhizoma Paris Mauritiae and Rhizoma Isatidis was closer and divided into one branch,while Rhizoma Paris in Zhuxi could be separately divided into another branch.Cluster analysis could be used to judge the relationship.
