Purpose:Geranium sibiricum L. is a kind of wonted Chinese drug. It has the function of eliminating the wind-damp,clearing the channels,and arresting diarrhea. It can be used in the symptoms of arthralgia due to wind-dampness, anaesthetization and muscular constriction,bones and muscles ache, and diarrhea in TCM. Geranium sibiricum L. compared with Geranium, Geranium sibiricum L. is rich in the northeast region. Sysrematic pharmacognostic study of Geranium sibiricum L. is carried out by chemical constituents, quality research and quality evaluation, thus lays the foundation for further developing new medical plants resources.Material and method:1. Distill Geranium sibiricum L.with ethanol, extract the part of ethyl acetate with silica gel column chromatography, gel column chromatography or recrystallization purification, according to its physical and chemical properties and the use of 1H-NMR , 13C-NMR and other spectroscopic methods, reference data reported in the literature, the structure of the obtained compounds were identified.2. Establish the methord of HPLC determination of Geranium sibiricum L. and determinate 5 contents of Geranium, including flavonoids and organic acids.Chose representative of the origin of plants, does a correlation analysis. And evaluate the ecological environment of 30 samples.3. Use inductively coupled plasma mass spectrometry (ICP-MS) detect the Geranium sibiricum L. herbs and soil content of 14 inorganic elements, and calculate the Zn / Cu, does the principal component analysis and cluster analysis to find the character,analysis the correlation of 14 elements in herbs, to find the disciplinarian between elements; and discuss thr relationship between herb and soil and analysis the constants of enrichment, provide the basis for the cultivation of medicinal herbs.4. Determination the contents of the active ingredient of 27 samples by HPLC, and 21 samples were selected to do principal component analysis and cluster analysis, finally selected out of 21 samples for similarity evaluation, established the ideal comparison chart and the fingerprints of Geranium sibiricum L.. 5. Analysis 10 different samples of Geranium sibiricum L. powder with X-ray diffraction, according to 2θangle positioning to similarity calculate and analysis with d/I/I0,and achieve the Fourier fingerprint of X-ray diffraction.Results and Conclusion:1. Isolated six compounds from Geranium sibiricum L., the compounds were identified: daucosterol (Ⅰ), kaempferol-3-O-rhamnoside (Ⅱ), kaempferol-7-O-rhamnoside (Ⅲ), kaempferol-3-7-O-α-L-II rhamnoside (Ⅳ), gallic acid (Ⅴ), quercetin (Ⅵ); The study will provide the experimental evidence for looking for reference material and further development and utilization of the medicinal resources.2. Establish the methord of HPLC determination gallic acid, protocatechuic acid, quercetin, kaempferol, kaempferol -3,7-O-α-rhamnoside five kinds of active ingredients of Geranium sibiricum L. the assay method is simplely operate, accurate, stable, and repeatable, principal component analysis of the comprehensive evaluation index can be directly reflects the quality of it.3. To do Principal component analysis of the data elements and determine the characteristics of herbs are Fe, V, Pb, As, Na, Hg, cluster analysis showed the content of inorganic elements of different regions can be divided; There was a significant correlation of inorganic elements in herbs and soil, indicating that Geranium sibiricum L. absorpt inorganic elements mainly is non-selective absorption, that is, passive absorption through the regulation of the content of inorganic elements in it. Geranium sibiricum L. have a strong ability in enrich of potassium, followed by theMg,Ni,Fe,V,Ga,Zn,Cu,and Zn/ Cu has a low point with 3.8. The results can be used as a chemical characteristics.4. Selected 27 samples and analyzed by principal component analysis and cluster analysis pattern recognition, and ultimately optimized 21 samples to do similarity evaluation, generate reasonable control spectrum5. Establish a new method of Geranium sibiricum L. of XRD analysis of fingerprint identification methord.
