Purpose:Through the He wei fan liu kang of bile reflux gastritis treated rats models,Observed the traditional Chinese medicine He wei fan liu kang may on bile reflux gastritis rats gastrointestinal and gastric mucosa tissue IKK/NF-kappa B/IL-1beta effects,To explore the possible mechanism of the He wei fan liu kang treatment of bile reflux gastritis.Material and method:50 SPF SD 7 ~ 8 weeks old healthy rats were randomly divided into 5 groups according to random number table method,respectively,the blank group,model group,Domperidone group,Aluminum phosphate gel group and He wei fan liu kang group,10 in each group.The 10 rats in the blank group were fed normally during the whole experiment,and the saline was treated as a placebo.In the molding stage,the remaining 40 rats were formed by gavage reflux fluid,once a day for 7 days.Entered the stage of treatment,in addition to the rest of the blank group daily at 9 a.m.40 rats by gastric reflux fluid in order to guarantee the stability of the model.After six hours,respectively,the TCM group was given He wei fan liu kang one lavage treatment,Domperidone group was given Domperidone liquor lavage treatment,Aluminum phosphate gel group was given Aluminum phosphate gel diluent lavage treatment,once a day,treatment for 28 days.In the treatment of phase calculated daily diet quantity and water quantity,and observe the spirit of rats and the colour change,before the experiment 1 and 8,15,22,29,35 days in the weighing in rats.At the end of the experiment,rats were dissected to determine the rate of gastric residues and small intestine propulsion.Western blot method was used to determine the expression of IKK?,NF-kappaB p65 and inflammatory factors(Il-1?)in gastric mucosa.Rt-pcr was used to detect the expression of IKK?mRNA,NF-kappaB p65 mRNA and inflammatory factors(Il-1?mRNA)in rat gastric mucosa.Observed the influence of the He wei fan liu kang to bile reflux gastritis model rats with gastric mucosa IKK? protein,NF-kappaB p65 protein,Il-1?protein and gastric mucosal cell IKK?mRNA,NF-kappaB p65 mRNA,Il-1?mRNA.Results: 1.In the first day before the experiment,there was no significant difference in the general state of rats in each group.7 days after the experiment,the blank group rats general state and the initial state is roughly same,but the rest of the group of rats are eating reduced the volume of water,weight growth is slow,dark color,laziness,loose stool,etc.A week after the treatment started,The results showed that the Domperidone group,Aluminum phosphate gel group and He wei fan liu kang group there was a significant improvement in the amount of body weight and diet in the group,which was gradually recovered,and the movement increased.After the experiment,the stomach of the rats was dissected.After the dissection,it was observed that:In the blank group,the gastric mucosa was clear and bright,and the gastric mucosa was ruddy and smooth,without bleeding or erosion.There was a small amount of yellowish-green mucous paste in the pylorus of the model group,and the gastric mucosa of the gastric antrum was slightly roughened,and there were exfoliation,accompanied by hyperemia,edema,erosion and appearance of bleeding points of different sizes.2.It was determine through gastric residues and gastric emptying.Compared with the blank group rats,the gastric residual rate of rats in the model group was significantly increased,and the small intestine propulsion rate decreased significantly,and the difference was statistically significant(P<0.05).It was found that that decrease in the gastric residual and the increase of the small intestine propulsive rate in the rat with the group of aluminum phosphate gel,the domperidone group and the He wei fa liu kang group were obviously(P<0.05).He wei fan liu kang and aluminum phosphate gel group differences statistically significant,after the comparison,He wei fan liu kang,the aluminum phosphate gel group reduced residual rate in stomach and small intestine motional rate increases more significantly(P<0.05),In addition,the differences in gastric residual rate and small intestine promotion rate were not obvious in the two groups of the polypanrione group and the aluminum phosphate group and the gastric reflux group(P>0.05).3.By Western Blot method,compared with the blank group,model group rats the distal gastric mucosa tissue IKK beta protein,the NF-kappa B p65 protein,IL-1 beta protein expression levels significantly higher than the blank control group,difference statistically difference(P<0.05);Compared with the model group,the Domperidone group,Aluminum phosphate gel group and He wei fan liu kang group's rats expression levels of IKK beta protein,NF-kappaB p65 protein,Il-1beta protein were significantly lower in gastric mucosal tissues of gastric antrum,and there were statistically significant differences(P<0.05).The expression of He wei fan liu kang group IKK beta protein expression and Domperidone group was significant(P<0.05).The NF-kappaB p65 protein,IL-1? protein expression was no significant difference in Domperidone group,Aluminum phosphate gel group and He wei fan liu kang group.4.By Rt-pcr method,compared with the blank group,model group rats the distal gastric mucosa tissue IKK beta gene,the NF-kappa B p65 gene,IL-1 beta gene significantly higher than the blank control group,the level of gene expression difference statistically difference(P<0.05);Compared with the model group,the Domperidone group,Aluminum phosphate gel group and He wei fan liu kang group's rats expression levels of IKK beta,NF-kappaB p65 and Il-1beta were significantly lower in the gastric mucosal tissue of the gastric antrum,and there were statistically significant differences(P<0.05).Domperidone group and He wei fan liu kang group IKK beta gene,the NF-kappa B p65,IL-1 beta gene expression compared with significant difference(P<0.05),compared with aluminum phosphate gel between way has no obvious difference(P>0.05).Conclusion:1.Through the general status and target detection,It is known that the rat model of bile reflux gastritis,which is prepared by using Sodium taurocholate,Pancreatic enzyme,lecithin,is relatively successful.2.The gastric mucosa has been repaired with the He wei fan liu kang.The He wei fan liu kang probably by reducing the experimental rat gastric mucosa tissues IKK/NF-kappa B/IL-1 beta protein and gene expression of inflammatory markers,inflammation to block the NF-kappa B pathway formed on gastric mucosa,It is one of the mechanisms He wei fan liu kang to treat bile reflux gastritis.3.It can promote Stomach-duodenum movement with He wei fan liu kang.In rats with gastric reflux,the rate of gastric refraction decreased,and the rate of small intestine increased,which proved that the gastro-duodenum movement could be effectively promoted by He wei fan liu kang.4.He wei fan liu kang is effective to treat bile reflux gastritis.In the process of treating bile reflux gastritis,He wei fan liu kang not only can promote stomach-duodenum movement,can protect the gastric mucosa,and standardization for the promotion of clinical application to provide the corresponding experimental basis.
