Purpose:1.in this thesis,we use the seeds of Plantago asiatica as the object of study,and optimize the salt baking technology of the seeds by orthogonal test.2.UV spectrophotometry was used to study the changes of monosaccharide composition before and after processing of Plantago asiatica.3.on the basis of the best processing technology of salt in the plantain seed,the quality standard of ten batches of Plantago asiatica was evaluated by HPLC.Material and method:1.Material Ten batch of Plantago and salted salted products from different habitats2.Method2.1 Orthogonal test method was used to optimize the salt-burning process of Psyllium seedlings,and the optimum processing technology of Psyllium seedlings was established.The temperature,processing time,brine concentration and saltwater volume were selected as the investigation factors.Three factors were designed,Chromatographic conditions: Column:Welch ultimate XB-18(4.6 mm × 250 mm,5 ?m);Detection wavelength: Geniposidic acid(254 nm);Verbascoside(322 nm),5% acetic acid in water(A + B = 100%);the flow rate was1.0 m L / min;the column temperature was 30 ° C;the injection volume was 20 ?L;the mobile phase A was methanol and the mobile phase B was 0.5%;Use of geniposidic acid,verbascoside glycosides as the evaluation index,the determination by high performance liquid chromatography.2.2 Pre-column derivatization-HPLC was used to determine the changes of monosaccharide composition before and after processing.2.3 The detection wavelength was: Geniposidic acid(254 nm,0 ~ 20 min);Verbascoside glycosides(322 nm,20-50 min);the flow rate was 1.0 m L / min;the column temperature was30 ° C;the injection volume was 20 ?L;the mobile phase A was methanol and the mobile phase B was 0.5% acetic acid in water + B = 100%),the mobile phase ultrasonic bubbles,gradient elution.The chromatographic conditions,geniposidic acid,verbascoside retention time was 17 min,44 min.The contents of geniposidic acid,verbascoside and isoverine glycosides in 10 batches of raw and salt sunbaked products were determined,and the quality standards of psyllium were studied.Results:1.Orthogonal experiment design L9(3 '),to determine the plantago salt Sunburn process;preferably Processed sub optimal process is: the best salt Sunburn process is to take the net plantago,processing temperature 160 a 180 C,there Buzzer,sprinkle aroma of 5%concentration of salt water spray,plus its weight 40%(mL / 100 g)of brine,processing 7 min,remove and let dry naturally,let cool.2.The content of mannose and xylose were increased,and the content of galactose and Arabia sugar decreased.3.The wavelength switching method was used to determine the contents of geniposidic acid,verbascoside and isogamidine before and after the batch processing of ten psyllium seedlings.The concentrations of geniposidic acid,verbascoside and isogamidine in the range of 0.007 ~350 ?g / m L,0.04 ~ 200 ?g / m L and 0.09 ~ 450 ?g / m L,respectively Good linear relationship.Conclusion:1.The experiment through orthogonal test,the car before the salt Sunburn process."People's Republic of China Pharmacopoeia" Processed norms Salt Sunburn psyllium,every 100 kg net psyllium salt 2 kg.The experimental selection of the car before the child,"the People's Republic of China Pharmacopoeia" contains two indicators of geniposidic acid and verbascoside,the result is more objective and accurate.The experimental and "People's Republic of China Pharmacopoeia" compared to the processing specifications more specific,clear,complete and comprehensive,for the plantago Zi Sunburn process optimization laid the theoretical foundation.2.The monosaccharide composition of plantain before and after salt processing has obviouschanges,and the effect of processing on monosaccharide should be considered in future research.3.From the selection of 10 different regions of psyllium products can be clearly seen in different areas of their geniposidase,verbascoside,abscisin and polysaccharides are different amounts.Therefore,in the study of psyllium quality control experiments,should take into account the origin,raw materials and salt Sunburn and other factors,so that the experimental results more convincing.
