Objective:To determine the HPLC fingerprint of Rhizome Polygonati.To optimize the quality standard of Rhizome Polygonati recorded of<The Chinese pharmacopoeia>.To optimize <Guizhou province Chinese herbal pieces processing specification>recorded the way of Rhizome Polygonati.As well as compares with other ways of the processed products. Methods: Adopted HPLC method to determine the fingerprint of Rhizome Polygonati.To analyse the fingerprint with similaritysoftware and clusetering method.To set up the qualitative and quantitative methods of Rhizome Polygonati and processed product with TLC.and UV.Increase the content of extract of items.On the basis of colour and lustre,extract,the content of prolysacharide and total saponins to come into being of comprenensive evaluation index,with factorial and orthogonal experiment to optimize the processeds.Compared the study to others to analyse the otherness. Results: Determined the HPLC fingerprint of Rhizome Polygonati,Its chromatographic conditions for adopting Hypersil ODS2 chromatographic column(5microns,250mm×4.6mm),acetonitrile-wateras mobile phase in gradient elution,flow rate of 1.2 ml/min,column temperature of 35℃, detection wavelength of 203 nm,sample amount 20μL.Build the TLC. of diosgenin,water soluble extractive,the concent of total saponins.Optimizeding the steamed is steamed 8h and bradied 12 h,Stirfrying with wine is steamed 4 times,steamed 3h and bradied 3h.The recorded of steamed is beyond others,the comprehensive score has significant difference(P<0.01). But the others have not significant difference(P>0.1).The highest of content of prolysaccharido and total sapoins is the expriment’s way,the highest of extract is “4 4 steam system”. The comprehensive score of stirfrying with wine has significant difference(P<0.01). The others have significant difference,too(P<0.01).The expriment is the second.The best colour and highest of extract is the expriment’s way.The highest of others is belonging to others.Conclusion: The way of the fingerprint is stable and feasible,the study provides scientific reference basis for processed Rhizome Polygonati.
