Trigonostemon thyrsoideum is one of the important members of the genus Trigonostemon,from Euphorbiaceae,which is mainly distributed in tropical and subtropical regions of Guizhou,Guangxi and Yunnan in China.It belongs to the unique folk medicinal plants in Guangxi.The research shows that the components of the T guenu plants is very rich including diterpenoids,coumarins,flavonoids,alkaloids,phenylpropanoids,and tannins,which have many kinds of biological activities such as bacteriostatic,anti-inflammatory,anti-viral,etc.The discovery of many novel structures Daphnane diterpenoids in T.thyrsoideum since 2000,T.thyrsoideum has gradually entered the sight of natural product researchers.Up to now,more than 40 new Daphnane diterpenoids have been discovered from this plant,which proves that it is one of the most eugenin-rich diterpenoids in the T.genus.Pharmacological studies have shown that T.thyrsoideum have strong anti-HIV-1 virus and anti-inflammatory pharmacological activities,etc,which proves that.thyrsoideum has high value in medicinal research.In this paper,the chemical constituents of T.thyrsoideum were studied in detail,in order to provide lead compounds with good biological activity for the development of new drugs.The total extract of the medicinal material extracted with 95%ethanol solution,extracted with petroleum ether,ethyl acetate and n-butanol reagent in succession to obtain three parts extract.After preliminary pharmacological activity screening,it was found that petroleum ether and ethyl acetate extract had strong cytotoxicity against human colon cancer HCT116 cell line,human cervical cancer HeLa cell line and human liver cancer HepG2 cell line,and n-butanol site dip The cream has weak cytotoxic activity only on human cervical cancer HeLa cell line.This test mainly separates the petroleum ether extract from the petroleum ether.It is separated by silica gel column chromatography and guided by UV-led,LC-MS and high performance liquid chromatography.After repeated silica gel column chromatography,gel column chromatography,MCI Separation and purification of the extract by column chromatography,medium pressure rapid ODS chromatography,preparative thin layer chromatography and semi-preparative high performance liquid chromatography.A total of 17 monomeric compounds were obtained Combined with high resolution mass spectrometry,nuclear magnetic spectroscopy data and literature comparison,were identified and are respectively:trigothysoid D(1),trigothysoid E(2),trigothysoid F(3),trigoxyphin K(4),trigochinin F(5),trigonothyrin E(6),trigonosin D(7),trigochinin A(8),trigochinin B(9),trigochinin D(10),trigochinin E(11),trigoheterin B(12),trigolin G(13),trigoxyphin A(14),(-)-loliolide(15),?-sitosterol(16),monopalmitin(17).Among them,the compounds Trigoxyphin K(4),trigoheterin B(12),and trigolin G(13),(-)-loliolide(15)were isolated from T.thyrsoideum for the first time.The target compound daphnane diterpenoids were quickly located by summarizing the UV spectral characteristics of the compound.Using a series of separation methods to separate the targets.A total of 11 daphnane diterpenoids(1,3-5 and 7-13)were obtained from Fr.12-13 Summarizing the literature data,we predicted the possible structures of nine unknown components,two of which may be new compounds.The inhibitory activity of compound(1-11,14 and 17)on HCT116 cell line,HeLa cell line and HepG2 cell line was determined by CCK-8 method.The results showed that compounds 1 and 2 had moderate proliferation inhibitory activity against three kinds of tumor cells.Compound 9 had moderate proliferation inhibitory activity against human cervical cancer HeLa and human liver cancer HepG2 cell lines with IC50 values of 5.96 and 14.74 ?g/mL,respectively.The proliferation inhibitory activities of compounds 1,2,and 9 on the three tumor cells were all in a concentration-dependent manner.
