Objective To observe the effect of Zishui Qinggan Decoction on blood pressure in spontaneously hypertensive rats(SHR),as well as the changes of aortic nuclear factor-kappa B p65(NF-kappa B p65),serum tumor necrosis factor-alpha(TNF-alpha),interleukin-6(IL-6),and to explore the inflammatory mechanism of Zishui Qinggan Decoction in reducing blood pressure.Methods Forty male SHR rats of SPF grade 13 weeks were divided into model group,high dose group(high dose group),low dose group(low dose group)and benazepril group according to the principle of complete random distribution of blood pressure.At the same time,10 WKY rats of the same strain and the same age were set as the normal group.The high dose group,the low dose group and the benazepril group were given corresponding drugs respectively,while the model group and the normal group were given distilled water.Blood pressure was measured once a week before and after administration for 8 weeks.Before and during administration,the general situation of rats was observed.After administration,the expression of NF-kappa B p65 in aorta was determined by Western blot,the expression of NF-kappa B p65 in aorta was determined by RT-PCR,and the levels of TNF-alpha and IL-6 in serum were determined by ELISA.Results (1)Blood pressure: SHR blood pressure was significantly higher than WKY rats.The systolic blood pressure of Zishui Qinggan Decoction group was significantly lower than that of Benazepril group(P < 0.01).There was no significant difference between the two groups before and after treatment,and blood pressure tended to be stable after 3 weeks of administration.The effect of each treatment group was similar(P > 0.05).(2)Level of NF-kappa Bp65 protein in aorta: The high-dose group of Zishui Qinggan Decoction and Benazepril group decreased significantly(P < 0.01).The high-dose group of Zishui Qinggan Decoction and Benazepril group had the same effect(P >0.05).(3)The level of NF-kappa Bp65 m RNA in aorta: the low-dose group,the high-dose group and the benazepril group of Zishui Qinggan Decoction plus or minus prescription all decreased,with significant difference(P < 0.01).The high-dose group and benazepril group of Zishui Qinggan Decoction plus or minus prescription had better effect(P < 0.05,P < 0.01).The effect of Zishui Qinggan Decoction plus or minus high dose group was similar to that of Benazepril group(P > 0.05).(4)Serum levels of TNF-a and IL-6: The serum levels of TNF-a and IL-6 in low-dose group,high-dose group and benazepril group of Zishui Qinggan Decoction were significantly decreased(P<0.01),and the effects of each treatment group were similar(P>0.05).Conclusion Zishui Qinggan Decoction can reduce blood pressure by lowering the levels of NF-kappa B p65,IL-6 and TNF-alpha,which confirms the possible mechanism of Zishui Qinggan Decoction from the inflammatory point of view.
