Objective: To investigate the effect of Zishuiqingganyin on blood pressure of SHR and the mechanism of blood pressure lowering.Methods: Forty SHR rats were randomly divided into SHR group,low-dose group,high-dose group and benazepril group,and 10 WKY rats were WKY group.The corresponding drug treatment was performed.Blood pressure was measured every week for 2 days,averaged and analyzed.After 8 weeks of intragastric administration,anesthesia,abdominal aorta blood collection and dissection of the thoracic aorta were performed for SOD,MDA,GSH serological examination and NOX4 protein of thoracic aorta.And mRNA testing.Result:1.Blood pressure: The blood pressure of each SHR rats before administration was statistically significant(P <0.01).There was no significant difference between the groups(P > 0.05).After 8 weeks of administration,compared with the SHR group,the blood pressure of the low dose group,the high dose group,and the benazepril group decreased,and there was statistical significance(P <0.01).There was no statistically significant difference between the three groups.2.Serum: The levels of SOD and GSH in the low-dose,high-dose and benazepril groups were higher than those in the SHR group(P <0.05),and the SOD activity was the best in the low-dose group(P < 0.01),GSH content in the high dose group is the most different(P <0.01);low dose group,high dose group and benazepril group can reduce serum MDA content,and The SHR group comparison(P <0.05)was effective in the low-dose group and the high-dose group(P <0.01).There was no significant difference between the three groups(P >0.05).3.NOX4 protein and mRNA: NOX4 protein expression in low-dose group,highdose group and benazepril group was significantly lower than that in model group(P <0.05),and high-dose group and benazepril group had better effect(P<0.05);the expression of NOX4 mRNA in each group was consistent with the expression of protein level.There was no significant difference between the two groups in high dose group and benazepril group(P > 0.05).Conclusion: Zishuiqingganyin can reduce the blood pressure of SHR rats,affect the concentration of SOD,MDA,GSH and the expression of NOX4 protein and mRNA,so it may affect the blood pressure level by regulating oxidative stress.
