1.PurposeThis study is to use intervention therapy with different frequency of electric acupuncture in patients with abdominal obesity,to observe the effect of electric acupuncture treatment on patients with intestinal flora,and the change of biochemical and simple body fat parameter index.To compare the effects of different electric acupuncture frequencies on intestinal flora and clinical effects in patients with abdominal obesity,and to provide appropriate reference for electric acupuncture intervention program optimization of abdominal obesity.2.MethodsClinical experiment:From September 2017 to November 2018,40 patients with abdominal obesity,aged between 18 and 50 years old,were admitted to the experiment,who were in treatment at the metabolism clinic of acupuncture department of Dongfang hospital at the Beijing University of Traditional Chinese Medicine.The patients were randomly divided into electro-acupuncture group ?(20 cases)and electro-acupuncture group?(20cases)by Excel random number grouping method.The two groups of patients were all treated with the principle of regulation of Dai Merdian,group I frequency selection(2 Hz),group II frequency selection(2/100 Hz)intervention.The intensity of abdominal muscles in patients was based on rhythmic contraction or slight trembling needle handle for degrees,average 2 v,with needle retained for 20 minutes.During the treatment,patients were asked to have more meals each day but less food at each time,and to avoid greasy and probiotic foods and medicines,and meanwhile recording exercise and diet with mobile phone AppDetection and analysis of intestinal flora:All stool samples collected from two groups of patients before and after electro-acupuncture treatment were sent to be tested.Data collected were analyzed by combination of clinical data of two groups of patients,so as to analyze the characteristics of intestinal flora in abdominal obesity patients and the changes of intestinal flora in two groups after electro-acupuncture intervention at different frequencies.3.ResultsAmong the 40 cases of abdominal obesity,39 cases were finally completed,including 20 cases in group I,19 cases in group II and 1 case was eliminated3.1 Analysis and comparison of intestinal flora:In terms of species distribution,high-throughput sequencing was applied to all samples before and after the intervention of electro-acupuncture at different frequencies contains:14 Phyla,25 Classes,40 Orders,68 Families,219 Genus and 445 Species,total OTU was 750.After electro-acupuncture intervention treatment,the proportion of Bacteroides in intestinal flora of the two groups,was higher than that of before treatment(P>0.05)in terms of the level of Phylum,Class and Order.Concerning the level of Phylum,the abundance of Firmicutes decreased and that of Bacteroidetes increased,But there was no significant difference(P>0.05).Regarding the Class level,the abundance of Clostridia decreased while that of Bacteroidia increased with no significant difference(P>0.05).The level of Order was similar to that of class.The abundance of Clostridiales decreased,while that of Bacteroidales increased3.2 Comparison of the body weight,waist circumference(WC),hip circumference(HC),body mass index(BMI),body fat percentage(BF%)and waist-hip ratio(WHR)of the two groups before and after treatment:After two courses of treatment,the weight,WC,HC,BMI,BF%and WHR of the two groups were improved in varying degrees.Among them,the weight,WC,HC,BMI and WHR of the two groups significantly improved(P<0.05)3.3 Comparison of curative effects between the two groups:After 2 courses of treatment in group I,6cases were cured,7cases markedly effective,2 cases effective and 5 cases not effective.After the same period of intervention in group ?,7cases were cured,6 cases markedly effective,2 cases effective and 4 cases ineffective.The overall effect in group II was 3.94%better than that of group ?.Compared with the total effective rate of the two groups,although the total effective rate of electro-acupuncture group ? is higher than that of electro-acupuncture group ?,(P>0.05)there is no significant difference in the total effective rate between electro-acupuncture group ? and electro-acupuncture group ?.The reduction of WC in group ? was better than that in group ?(P<0.05)4.Conclusion4.1 On the basis of no statistical difference in sex,age,abdominal obesity related indicators and blood lipids between electro-acupuncture group I and electro-acupuncture group ?,the frequency of(2Hz)electro-acupuncture and(2/100Hz)electro-acupuncture both can reduce weight and improve clinical symptoms of abdominal obesity patients.The curative effect is similar,and(2/100Hz)electro-acupuncture is better for waist circumference reduction4.2 Intestinal flora is closely related to abdominal obesity.After electro-acupuncture intervention,it was found that the abundance of Bacteroides in intestinal flora of abdominal obesity patients increased at different levels,suggesting that the regulation of intestinal flora in the treatment of abdominal obesity patients by electro-acupuncture intervention may be related to weight loss and waist loss,and that intestinal flora imbalance may be one of the reasons affecting the occurrence and development of abdominal obesity.
