Objective: By observing the effect of the acupuncture method of "guben peiyuan,Jianpi hewei" on the body weight,blood glucose,food intake,and related indexes of brain-gut peptide in rats with high-fat diet induced simple obesity,combined with the results of previous studies on intestinal flora,the effect of acupuncture on intestinal flora in obese rats was explored through the "bacteria-enteric-brain" axis,so as to provide in-depth theoretical support for the clinical treatment of simple obesity with acupuncture.Methods: The rat model of simple obesity induced by high fat diet was prepared.8 rats were randomly selected from the model group,8 from the acupuncture group,and 8 from the western medicine group according to the random principle,and 8 rats from the normal diet were given as the control group.The acupuncture group and the western medicine group were treated for 4 weeks.During the treatment,no changes were made in the feed of each group.The acupuncture group was treated with the acupuncture method of "guben peiyuan,Jianpi hewei".Western medicine group was treated with metformin hydrochloride sustained release tablets by gavage.During the intervention period,the changes of survival status,food intake and body weight of rats in each group were observed regularly.Rats were sacrificed for sampling after 4 weeks of intervention.The fat weights of epididymis,perirenal and perienteral of rats in each group were measured.ELISA was used to measure the contents of serum peptides in rats.Results:(1)According to the previous study,the species difference of intestinal flora of rats in the model group was greater than that in the control group and the treatment group,and both the abundance of intestinal flora and the mean distribution of species were lower than those in other groups.After intervention,the content of beneficial bacteria in the intestinal tract of rats in the treatment group was higher than that in the model group,among which,the proportion of bacteroidetes increased and the proportion of firmicutes decreased significantly.(2)Survival state of rats : control group rats living condition is good,the feeding,drinking water,urine normal,luxuriant hair shiny,sensitive,high degree of response to external stimuli.Model group rats with poor living conditions,feeding,water adding,defecate to powder is faulty,quantity the smell of urine,more lie without god,matt dark hair and wool,low degree of response to external stimuli.After intervention,the survival status of rats in the acupuncture group and the western medicine group(drinking water,defecation,hair,mental state,etc.)was improved better than that in the model group,and the rats in the acupuncture group had more significant improvements in drinking water,stool,hair and mental state than those in the western medicine group.(3)The body weight,peripheral fasting blood glucose and visceral fat wet weight of rats in the model group were significantly higher than those in the control group(P<0.05).After intervention,the body weight,peripheral fasting blood glucose and visceral fat wet weight of rats in the acupuncture group and western medicine group were significantly reduced.The acupuncture group was significantly lower than the western medicine group(P<0.05).(4)There was no significant difference in the food intake at each time point between the model group and the control group(P> 0.05).There was no significant difference in the food intake between the acupuncture group and the western medicine group at each time point(P> 0.05),but the acupuncture group and the western medicine group had no significant difference.Rat food intake showed a steady downward trend;The food intake of the model group rats was significantly higher than that of the control group(P <0.05).After intervention treatment,the food intake of the acupuncture group and the western medicine group was significantly decreased(P <0.05),and the acupuncture group was significantly lower than the western medicine group.The decrease was significant(P <0.05).(5)The serum levels of Ghrelin,PYY,GLP-1,and CKK in the model group were significantly lower than those in the control group(P <0.05).After intervention,the serum levels of Ghrelin,PYY,GLP-1,and CKK in the acupuncture group and the western medicine group were significantly increased,and the acupuncture group was significantly higher than that of the western medicine group(P <0.05).Conclusion:(1)Rats with simple obesity induced by high-fat diet had poor survival status,overweight,increased food intake,blood glucose,visceral fat,intestinal flora and other disorders.(2)"Guben Peiyuan,Jianpi hewei " acupuncture can benignly regulate the structure of disordered intestinal flora,increase the number of Bacteroides and beneficial bacteria(Bifidobacterium,Lactobacillus,etc.),and make thick Decreased flora.(3)"Gubenpeiyuan,Jianpi hewei " acupuncture and metformin hydrochloride sustained release tablets can significantly improve the survival of obese rats.Both of them can effectively reduce the weight and peripheral fasting blood glucose of obese rats.Both of them can effectively reduce the food intake and visceral fat weight of obese rats.All of them can significantly increase the contents of Ghrelin,PYY,GLP-1 and CKK in serum of obese rats,and exert the effects of weight loss,hypoglycemia,lipid-lowering and food inhibition by acting on the central nervous system through the enteroencephalon axis.Acupuncture is more effective than metformin hydrochloride sustained release tablets.(4)"Guben Peiyuan,Jianpi hewei " acupuncture is reasonable and effective in treating obesity in acupuncture,which can provide experimental basis for clinical acupuncture treatment of obesity.
