Objectives:This study aims to refine the empirical prescription of professor kong guangyi for the treatment of type 2 diabetes under the guidance of his shaoyang trifocal membrane system theory,and at the same time explore the extraction method of the empirical prescription,which is one of the important contents inherited by famous TCM doctors,so as to make it scientific and standardized.Research on the clinical use of auxiliary platform's collection of prescription were analyzed,and the computing experience in the treatment of type 2 diabetes at the core of the drug,and further with the aid of network pharmacology,adopt the method of simulating research ingredients-targets-multidimensional and multifaceted relationship between disease and explain its mechanism of action,to verify the experience of drugs and curative effect,finally under the guidance of traditional Chinese medical theory to establish experience.Methods:Firstly,the outpatient cases and prescriptions of type 2 diabetes diagnosed and treated by professor kong between 2010 and 2015 were reviewed,and TCMISS was recorded.With the help of the frequency statistical function of the platform,this paper explored the commonly used traditional Chinese medicine and its four qi and five flavors and commonly used dose of kong guangyi in the treatment of type 2 diabetes.With the help of the platform's key drug calculation,association rule calculation and other functions,explore the core drug and drug combination,combined with the network visual display,and extract the empirical formula.Then through TCMSP to find out the chemical components of the drug in the experiential prescription and its target,and with the help of Inparanoid complete gene mapping,establish the experiential target database;At the same time,HPRD data was used as the research background network database.Combined with the known target of type 2 diabetes retrieved in DisGeNET,the potential target of type 2 diabetes was found and the target database of type 2 diabetes was established.CytoScape was used to find the drug-acting protein,enrichment analysis was performed on DAVID and Kobas to find the key protein and signal pathway,and discuss the action mechanism of the empirical side.Results:A total of 41 cases of type 2 diabetes were collected.The age distribution of the patients was approximately normal.Commonly used traditional Chinese medicine radix ophiopogonis,radix paeoniae rubra,Fried atractylodes,cortex phellodendri,salvia miltiorrhiza,radix scutellariae,radix glycyrrhizae,radix trichosanthis,rhizoma anemarrhenae,radix paeoniae alba,pinellia,ginseng,radix achyranthis bidentatae,turmeric,cinnamon,poria cocos,including cold LiangYao accounted for 52%,warm medicine accounted for 33%,bitter medicine accounted for 42%,adj uvant(32%),bitter herbs(19%).The core drugs and their usual dosage were 30g radix ophiopogonis,30g salvia miltiorrhiza,lOg radix paeoniae rubra,lOg radix scutellariae,15g cortex phellodendron,and 10g radix atractylodis.By searching TCMSP,202 molecules of traditional Chinese medicine were obtained,including salvia miltiorrhiza,radix ophiopogonis,radix paeoniae rubra,radix scutellariae,radix huangbai,radix atractylodis,and radix atractylodis.A total of 1279 diabetes-related target proteins were obtained by DisGeNET,and 4869 potential target proteins of type 2 diabetes were obtained by binding HPRD.Finally,168 target proteins were obtained.DAVID was used for enrichment analysis Cluster 1 had the highest enrichment value(4.41),including Rapl signaling pathway,proteoglycan in cancer,and focal adhesion.The group with the highest association with type 2 diabetes was cluster 5,with an enrichment value of 2.97.The PI3K/Akt signaling pathway was added after the addition of Kobas database.There are 26 key targets:PKLR?IKKB?MAPK1?TNF?PI3KCG?INSR?MTOR?ACACA?GSK3??FASN?CALMI?PYGM?IL6?MET?EGFR?RXRA?EGF?COL1AI1?KDR?BCL2?IL2?TP53?HSP90?VEGFA?ITGB3?Conclusions:Under the guidance of the sanjiao membrance system theory,type 2 diabetes should be caused by pathogenic factors.Therapeutic drugs are:with radix ophiopogonis,radix Scutellariae,radix trichosanthis,rhizoma anemarrhenae,represented by nourishing Yin clearing lung heat,represented by salvia miltiorrhiza and radix paeoniae rubra,radix curcumae,radix paeoniae alba,cold blood,blood to Fried atractylodes,ginseng,tuckahoe,liquorice,dried tangerine or orange peel,pinellia tuber of spleen clearing damp phlegm,cortex phellodendri,cinnamon,radix achyranthis bidentatae,cortex eucommiae,fructus alpiniae oxyphyllae,amomum villosum represented by latent Yang removing spleen and kidney;Professor kong guangyi's prescription for the treatment of type 2 diabetes should be 30g radix ophiopogonis,30g salvia miltiorrhiza,lOg radix paeoniae rubra,lOg radix scutellariae,15g radix huangbai and lOg radix atractylodis.According to the research on network pharmacology,the empirical prescription may treat type 2 diabetes mainly by regulating the PI3K/AKT signaling pathway.
