At the same time,the improvement of people's living standard has also brought a change in lifestyle,the incidence of diabetes,the effects of lifestyle and genetic factors are all lead to DM,in the truth,95%of patients with diabetes is T2DM,with its exponential growth,it will become the world's serious health problems,diabetes patients in China is also increasing.The amount of population and a considerable part of the glucose tolerance,increased the reserve force for Chinese T2DM patients.Traditional Chinese medicine syndrome with the unique charactors of differentiation and quantitative compatibility,it has played an important role in the treatment of diabetes,but the traditional Chinese medicine compound complex composition,the mechanism is still not clear,with multiple components,multiple targets and system control theory,complex network model is set up based on the exploration of drug protein network pathway.To find out the drug of multi targets,drug molecules and protein interaction of research strategy from the target in the body,in study of Chinese medicine treatment of diabetes is feasible.1.Research objectiveThe retrieval strategy was as follows:CNKI,Wanfang and VIP were retrieved with search terms as diabetes,experiment,T2DM,in order to obtain the relative documents in taditional Chinese herb;China's traditional Chinese medicine(TCM)chemical composition database,Drugbank and Chinese Pharmacopoeia"(2010 version)were retrieved with TCM chemical composition,in order to obtain the information of Chinese herb,components and trgets to shape the data set.Build the complex network model of Formula-Component-Target,the relationship of the model was evaluated by Cytoscape,to explore traditional Chinese medicine function mechanism in the treatment of DM,provide the reference for the target specific drug research,so as to broaden the new mode of drug development,shorten the time for drug development;research trend followed by multiple components,multiple targets multi path,also provides the reference for the experimental study.2.Methods and contents(1)In the study of diabetes as the research object,the retrieval test class related to Chinese medicine treatment of diabetes in literature CNKI,Wan Fang,VIP database,through the integration of the relevant information,based on the literature of the "DM-Chinese medicine"and "component targets","traditional Chinese medicine composition target dataset;(2)Using Cytoscape software as the carrier,the "DM-" component,the ingredients of Chinese medicine "target","traditional Chinese medicine composition target dataset visualization analysis,the establishment of natural drug compounds(Chinese Medicine)-target(path)of complex networks.The principle of the connection between the nodes is a compound that is produced by a certain target or by a certain path,which is the same as that of natural medicine.At the same time,the whole network is described and analyzed.(3)The complex network method was used to study the drug target interaction network.The network parameters of drug target interaction network were calculated and analyzed,and the power-law index of drug target interaction was found.(4)Compound used for the treatment of diabetes prescription or results have been clear to verify the model.Take the traditional Chinese medicine contained in the compound as the target node,select these points in the network model,and extract the sub networks which are related to these nodes.Objective to analyze the feasibility of the target information in the sub network for the intervention of diabetes.3.Research resultsThis study is based on the traditional Chinese medicine pharmacology in type 2 diabetes as the source of literature in laboratory animal,The Formula-Component-Target DS involves a total of 149 types of herbs such as ginseng,astragalus membranaceus,ligusticum wallichii,fuling,white atractylodes rhizome.Covers the common prescription of diabetes.It shows that the improvements of the blood glucose,blood fat,the function of pancreas,the release of inflammatory factor and the modulation of brain central nervous was related to the targets effects of the Chinese herb and its effective components.The Formula-Component-Target model is a multiple-link,multi-approach and small-world network.The network node of the study is concentrated,which shows the tightness of the model.Finally,based on the cmmonly used prescription,Xiaoke recipe decoction,Rehmannia six,seven kinds oof powder such as clinical treatment of type 2 diabetes,do accurate model test,which included kudzuvine root,Coptis chinensis,Fructus Gardeniae,Radix Scrophulariae,astragalus,Poria,Radix Aucklandiae,etc in medicine and prescription of traditional Chinese Medicine model,component and the target information of the fit,the constructed network reaction target pathway of this model is part of the drug.4.DiscussWith the continuous development of social economy,people's life style has also been a huge change,which makes the proportion of patients with diabetes increased significantly,has become one of the major diseases endangering people's health.The traditional Chinese medicine in TCM theory and dialectical theory of governance under the guidance of the quantitative way and play a role,however,traditional Chinese medicine compound due to its complex composition,complex process and unique philosophy system,it is difficult to be recognized by patients and the medical profession in the world and understand.The emergence of complex networks,starting from the components and targets,and the construction of the complex network model with the modern visualization research methods,have an important role in promoting the development of traditional Chinese medicine.Starting from the perspective of complex network to construct,network properties and network connectivity analysis,clustering coefficient and EAGLE clustering analysis of traditional Chinese medicine and method for diabetes component target network,network components,components of Chinese Medicine-Chinese medicine network model,the target component of the traditional Chinese medicine point network as an example the results show that the network is a multi way,multi link small world network,and part of the node degree is relatively low,combined with pharmacological research and analysis,the effective components of Chinese medicine more,including quercetin,curcumin and berber:ine effects are more,and repeat target of three drugs less;ginseng,Poria,Herba Hyperici japonici,Wu Zhuyu a traditional Chinese medicine containing traditional Chinese medicine component is the most effect,the most.To sum up,that target components of traditional Chinese Medicine-point network with scale-free property;and the network clustering results show that at least every Chinese medicine there is a relationship between the active ingredient and the target that Chinese medicine is an effective ingredient can regulate target related role in treatment of diabetes.Research on the network target of Chinese medicine is an important breakthrough in modern medical research.The three networks are small world networks,the data in the existing research is far from enough,and the complex relationship between the effective components,the target,the role of the pathway has not been clarified.For more complex network model in the research process,also need to boldly explore and use more appropriate network principle,suitable for the development of modern Chinese medicine to learn from the existing part of modern medical technology and tools in the analysis of the mode of interaction between the effective components and targets and associated targets closer to the operation,limitation break through the existing research.
