Intervention Effect Of Different Processed Rhubarb Products On Early Diabetic Nephropathy Rats

Objective To investigate the protective effects of different processed rhubarb products(raw rhubarb,wine rhubarb,cooked rhubarb and rhubarb charcoal)on early diabetic nephropathy rats.Methods 150 male rats were randomly divided into normal group and modeling group.After successful modeling,the modeling group was randomly divided into model group,telmisartan group,raw rhubarb high,middle and low group,ripe rhubarb high,middle and low group,wine rhubarb high,middle and low group,rhubarb charcoal high,middle and low group.In the modeling group,1%streptozotocin solution was intraperitoneally injected into each rat.The dosage of telmisartan group was 30 mg/kg/d.The dosage of four Processed Rhubarb products in high,middle and low groups was 4.164 g/(kg.d),2.083 g/(kg.d),1.042 g/(kg.d),respectively.The normal group and model group were given distilled water 2 ml/day,once a day for 8 weeks.12 hours after the last administration,samples were taken after anesthesia.Detection indicators are as follows: 1.Blood samples were taken to test random blood sugar before and after treatment;2.Detection of microproteinuria at the 2nd,4th,6th and 8th weeks to determine whether the disease is alleviated;3.Abdominal aorta blood samples were taken to test urea nitrogen and serum creatinine to observe whether the disease is improved;4.Measurement of kidney weight to detect whether the kidney is enlarged;5.HE staining to observe the kidney structure of diabetic nephropathy rats.Pathological changes;6.Immunohistochemistry was used to detect the expression of growth transforming factor-?1(TGF-?1).Result 1.Effect on blood sugar in rats: Compared with before and after treatment,the high-dose group of raw rhubarb,each dose group of wine rhubarb and each dose group of riperhubarb all decreased(P < 0.001);Compared with model group,the dose groups of telmisartan,wine rhubarb,ripe rhubarb and raw rhubarb all dose groups decreased(P< 0.05);Compared with telmisartan group,the dose groups of wine rhubarb and ripe rhubarb all dose groups decreased(P < 0.05).The middle and high dose groups of raw rhubarb all decreased(P < 0.05).2.The effects of Telmisartan group,high-dose cooked rhubarb group,raw rhubarb group and wine rhubarb group on microalbuminuria in rats: compared with model group,the contents of urinary albuminuria decreased 4 times(P < 0.05)and low-dose rhubarb group(6th,8th weeks);medium-dose rhubarb group(4th,6th,8th weeks);and rhubarb charcoal group(4th,6th,8th weeks)all decreased(P < 0.05).Compared with telmisartan group,microproteinuria decreased in low dose group,medium dose group(2nd,8th week),high dose group(8th week),low dose group(2nd,4th,6th week),medium dose group(2nd,4th week),high dose group(2nd week),low dose group(6th,8th week)and high dose group(P < 0.05).);3.Effects of creatinine(CRE)and urea nitrogen(BUN)on serum creatinine(CRE)and urea nitrogen(BUN)in rats: Compared with the model group,telmisartan,wine rhubarb,ripe rhubarb and raw rhubarb groups all decreased(P < 0.05);compared with telmisartan group,wine rhubarb and ripe rhubarb groups all decreased(P < 0.05),while raw rhubarb and rhubarb charcoal groups were higher(P < 0.05).CRE results showed that compared with the model group,telmisartan group,wine rhubarb group,ripe rhubarb group and raw rhubarb group all decreased(P < 0.05);compared with telmisartan group,wine rhubarb group,ripe rhubarb group and raw rhubarb high dose group all decreased(P < 0.05),while raw rhubarb medium-low dose group and rhubarb charcoal group were higher(P <0.05).4.The effects on kidney weight of rats: Compared with model group,telmisartan group,wine rhubarb group,ripe rhubarb group and raw rhubarb group all decreased(P < 0.05);compared with telmisartan group,wine rhubarb group,ripe rhubarb group and raw rhubarb high dose group all decreased(P < 0.05),while raw rhubarb medium and low dose,rhubarb charcoal group were heavier(P < 0.05).);5.Histopathological changes of kidney tissues of rats in each group: HE staining results showed that glomerulus enlarged,mesangial area widened,renal capsule narrowed,surroundingrenal tubular cells degenerated and lumen narrowed in model group;glomerulus narrowed,mesangial area narrowed,renal capsule widened,peripheral renal tubular degeneration alleviated and lumen enlarged in each group except rhubarb charcoal group;6.Influences on the expression of TGF-?1 in kidney tissues:Immunohistochemical method was used to detect the expression of TGF-?1 in kidney tissues: Compared with model group,the number of positive cells in different dose groups of raw rhubarb,ripe rhubarb,wine rhubarb and charcoal of rhubarb decreased(P < 0.05);Compared with blank group,the number of positive cells in raw rhubarb,ripe rhubarb,wine rhubarb and wine rhubarb decreased(P < 0.05).The number of positive cells in each dose group of rhubarb charcoal was still high(P <0.05).Conclusion 1.Wine rhubarb,ripe rhubarb and raw rhubarb can reduce the random blood sugar of DN model rats,and the effect of high-dose wine rhubarb is the best;raw rhubarb,ripe rhubarb,wine rhubarb and rhubarb charcoal can reduce the microproteinuria,urea nitrogen and creatinine of DN model rats,the effect of wine rhubarb is better than that of ripe rhubarb,followed by raw rhubarb and rhubarb charcoal.Secondly,the effect of high-dose Rhubarb wine is the best;2.Rhubarb wine,raw rhubarb,ripe rhubarb,rhubarb charcoal can reduce the expression level of TGF-?1 protein factor in kidney of DN model rats,play a role in delaying the development of early diabetic nephropathy and protecting kidney.
