Objective: To induce the formation of hyperlipidemia in TLR4 knocko ut mice by high-fat feeding,to explore the relationship between Huxin kang and Wnt signaling pathway,and the effect of Huxinkang on Wnt-1 and DVL gene expression in Wnt signaling pathway.Methods: 70 TLR4 knockout mice were divided into a blank control group and a hyperlipidemia model group according to the weight and sex of the mice.After successful modeling,they were divided into mo del group,medium and high dose Huxinkang group,atorvastatin calciu m tablet group and Wnt inhibitor WIF group according to random nu mber table method.Ten wild mice were normal control groups.The t wo control groups were fed with normal irradiation feed,and the rest were fed with high-fat diet(containing 21% fat,0.15% cholesterol),and the modeling time was 14 weeks.After the model was successfully ad ministered,the doses of the middle and high care HuXinkang group w ere 2.5g/(kg·d),5g/(kg·d),at the same time,intraperitoneal injection of physiological salt 0.3ml/d,once/d,WIF group for Wnt inhibitor intrap eritoneal injection,Wnt inhibitor XAV939,according to 2ug,intraperit oneal injection,diluted with saline,0.3ml/d,once/d,At the same time,distilled water was administered to the stomach,0.3ml/d.In the wild control and blank group,0.5ml of distilled water was intragastrically administered,and 0.3ml of normal saline was intraperitoneally injected once a day.Each group was administered continuously for 4 weeks.After 16 weeks,the mice were sacrificed and blood lipids were taken from the eyeballs of the mice.The levels of Wnt-1 and DVL in bloo d were detected by enzyme-linked immunosorbent assay(ELISA),and the expression of Wnt-1 and DVL genes was detected by real-time flu orescent quantitative PCR.Results:1.Effects on biochemical indicator:The levels of cholesterol and triglyceride in the model group were significantly higher than those in the blank control group(P<0.05);compared with the model group,atorv astatin group,high and medium doses The levels of cholesterol and tri glyceride were significantly lower in the group(P<0.05),and the effect of Wnt inhibitors on lowering blood lipids was not obviou s.There was no statistical difference in the effects of three drugs on high-dens ity lipoprotein and low-density lipoprotein.The significance of learnin g(P>0.05),the effect is not obvious.2.The effect of Wnt-1 and Dvl levels in blood:The levels of Wnt-1 and DVL in the model group were significantly higher than those i n the blank control group(P<0.05).The levels of Wnt-1 and DVL in the model group were higher,indicating hyperlipemia.Compared with t he model group,atorvastatin,Huxinkang and Wnt inhibitors all reduced Wnt-1 and DVL,and the difference was statistically significant(P<0.05);Wnt Inhibitors in the WIF group had a stronger effect on Wnt-1 a nd Dvl,and the difference was statistically significant(P<0.05).There was no significant difference betwe en the high and middle doses of H uXinkang group and atorvastatin group(P>0.05).3.Effect on Wnt-1 and DVL gene expression:The gene expression of Wnt-1 and DVL in the model group was significantly higher than t hat in the blank control group(P<0.05).The expression levels of Wnt-1 and DVL genes in the model group were higher,indicating that hyp erlipidemia and Wnt signaling pathway existed.Close association;comp ared with the model group,atorvastatin,Huxinkang,Wnt inhibitors all re duced Wnt-1,DVL,the difference was statistically significant(P<0.05);Wnt inhibitor WIF group reduced Wnt-1,DVL had a strong effect,the difference was statistically significant(P<0.05);there was no significan t difference between the high and middle doses of Huxinkang group a nd atorvastatin group(P>0.05).Conclusion:1.Huxinkang can reduce blood lipid levels.2.Huxinkang can significantly reduce the expression of W nt-1 and DVL genes,and its anti-hyperlipidemic effect may be related to this.
