Objective:To investigate the relationship between atherosclerosis and Wnt signaling pathway in Apo E-/-mouse atherosclerosis model induced by high-fat diet and the effect of Huxin Kang on the expression of Wnt signaling pathway.Methods:C57BL/6 mice were used as wild control group.All the animals were fed with Apo E-/-mice.The SPF level was suitable for 1 week.The rats were randomly divided into normal control group?n=8?and model group?n=52?.The normal control group was fed with normal diet.The model group was fed with high fat diet?including 21%fat and 0.15%cholesterol?according to the literature[10].After 13 weeks of feeding,four thoracic aorta were randomly selected from the model group,The mice were sacrificed in the course of modeling.Eight mice were killed in the course of modeling.They were divided into high,medium and low dose Huxinkang group,atorvastatin group and model group.The rats in the control group and the model group were treated with normal saline,and the rats in the control group and the model group were treated with normal saline.The high,medium and low dose of Huxinkang group were treated with 86.4g/?kg·d?,43.2g/?kg·d?,21.6 g/?kg·d?,the atorvastatin group was given3mg/?kg.d?,0.5ml/time/day for 4 weeks.At the end of 17 weeks,the mice were sacrificed and the thoracic aorta was taken to observe the plaque.The collagen content was measured and the expression of Wnt-1 and APC protein was detected by immunohistochemistry.Result:?1?The effect of plaque morphology:Compared with the model group,the high and middle dose of care group and atorvastatin group can significantly reduce the plaque area,reduce the degree of plaque lesions,the difference was statistically significant?P<0.05?.There was no significant difference between the two groups?P>0.05?.Compared with the middle dose Huxinkang group,the high dose of Huxinkang group could reduce the plaque area,?P<0.05?.There was no significant difference between the two groups?P>0.05?.Compared with atorvastatin group,there was no significant difference between the two groups?P<0.05?,Dosage care group was able to reduce plaque area,which was statistically significant?P<0.05?.HE staining showed that there was no obvious plaque formation in the control group,the aortic wall was clear,the intima was smooth and the endothelial cells were neatly arranged,and the smooth muscle cells were well shaped.In the model group,a large number of plaques were formed,a large number of lipid deposits were observed in the endometrium,and the thickening of the intima was observed.A large number of irregular foam cellsandsmoothmusclecellswerearrangedintheplaque.Drug intervention group also showed a greater degree of intimal thickening,compared with the model group,plaque lesions reduced,plaque area smaller,reduced foam cells.Pathological changes:no obvious plaque formation in the blank control group,the wall of the aortic wall was clear,the intima was smooth and smooth,the endothelial cells were arranged neatly and the smooth muscle cells were well shaped.In the model group,a large number of plaques were formed,a large number of lipid deposits were observed in the endometrium,and the thickening of the intima was observed.A large number of irregular foam cells and smooth muscle cells were arranged in the plaque.Drug intervention group also showed partial thickening of the endometrium,a small amount of plaque formation,smooth muscle cell arrangement is still neat,compared with the model group,plaque lesions reduced,plaque area smaller,reduced foam cells.?2?Comparison of collagen content:Masson staining showed that collagen fibers were increased,thickened and arranged in model group and drug intervention group compared with the control group.Compared with model group,collagen fibers in drug intervention group Reduce the arrangement is still neat,compared with the atorvastatin group,high-dose care Huxinkang group of collagen fibers is relatively reduced,arranged yet,low-dose care Huxinkang group collagen fibers slightly more.?3?Effects on Wnt-1 and APC protein expression:The positive expression of Wnt-1 and APC protein was observed in the cytoplasm of the cells.The more brown-yellow granules were,the more obvious the expression was,and the control group showed no brown The expression of Wnt-1 and APC protein in the model group was significantly higher than that in the control group?P<0.05?.The expression of Wnt-1 and APC protein in the model group was significantly higher than that in the model group The expression of Wnt-1 and APC in statin group was significantly lower than that in statin group?P<0.01?.There was no significant difference in the expression of Wnt-1 and APC between the low dose group and the model group?P>0.05?.Compared with the statin group,the expression of Wnt-1 and APC in the high dose of Huxinkang group was statistically significant?P<0.05?.There was no significant difference between the two groups?P>0.05?.Conclusion:?1?The high expression of Wnt-1 and APC in atherosclerosis in Wnt signaling pathway confirmed that there was a certain relationship between atherosclerosis
