Objective: To observe the anti atherosclerosis effect of Huxinkang tablets and the influence of inflammatory factor IL-1 beta,IL-6 alpha and TNF-?,effects on NF-?B signaling pathway expression?To elucidate its mechanism,new ideas to find new and effective method for the prevention and treatment of atherosclerosis with traditional Chinese medicine.Methods: 78 healthy SD rats were randomly divided into control group(n = 13).The model rats were injected with large doses of vitamin D3 and high-fat diet for 12 weeks,thirteenth weeks,6 rats were randomly selected from the abdominal aorta,oil red O staining section to observe whether the formation of atherosclerotic plaque,prove the success of the atherosclerosis model.The rats in the model group were divided into five groups: the model group,the positive control group,the high dose group,the middle dose group and the low dose group.The model group with distilled water 10ml/kg;low dose group,middle dose group,high dose groups were 0.41g/kg/d,0.82g/kg/d,1.64g/kg/d dose,the positive control group with atorvastatin calcium administration by 2mg/kg/d,the drug was diluted by distilled water intragastric gavage volume is 10ml/kg.After 12 weeks of treatment,abdominal aorta by oil red O staining under the microscope;double antibody sandwich enzyme linked immunosorbent assay(ELISA)method was used to detect the serum inflammatory factor IL-1?and IL-6 and TNF-?expression level;the arterial tissue by protein immunoblotting(Western blotting)expression level detection method NF-?B signaling pathway protein analysis system,the gray value of image analysis,the ratio of protein and ?-acting reference protein expressed in tissue protein expression of NF-?B.Result:1.General situationThe control diet group rats were normal,sensitive reaction,hair colorgloss neat,rats were given intraperitoneal injection of vitamin D3 injection,appear emaciation,eating less,dull coat,hair loss,lack of energy,slow reaction etc..Group after administration,administration of rats body weight increased significantly,the hair color to restore luster,sensitive reaction gradually,the model group high dose group,middle dose group and positive drug group rats weight growth comparison,the difference was statistically significant(p<0.05),compared with the low dose group,no significant difference(p>0.05).2 Aortic morphological changesAfter oil red O staining visible: the intima of AS rats of model group were significantly thickened,atherosclerotic plaque in red staining(lipid droplets),low dose group on the inner wall of the lipid droplets compared to the model group had no obvious change,but the high dose group of aortic vascular smooth,no obvious atherosclerotic plaque.3.Effect of on inflammatory factors in serum of ratsCompared with the model group,positive control group and Huxinkang tablets in low dose group and middle dose group and high dose group serum IL-1?,IL-6 and inflammatory factors were decreased,and the middle dose group,high dose group,positive control group were compared,significant difference was statistically significant,high dose of Huxinkang compared with the positive control group,IL-1?,IL-6 level decreased,but the difference was not statistically significant(p>0.05),TNF-?content was significantly decreased,the difference was statistically significant(p<0.05).There was no significant difference between the model group and the low dose group.4 Effects of B on the expression of NF-?B signal pathway in rat aortaCompared with the model group,positive control group and Huxinkang tablet low dose group,middle dosage group and the NF-?B signaling pathway protein in aortic tissue of high dose group the expression levels were decreased,and the middle dose group,high dose group,positive control group were compared,difference was statistically significant(p<0.05).However,there was no significant difference between the model group and the low-dose group(p>0.05).Conclusion:1 Huxinkang Tablet can obviously reduce the formation of atherosclerotic plaque and improve the symptoms of AS.2 Huxinkang by expression of NF-?B signaling pathway protein,blocking the activation of this signaling pathway,thereby inhibiting rat inflammatory cytokine of IL-1?,IL-6 and TNF-?release,achieve the anti atherosclerosis effect.
