Objective:This study was to investigate the effect of acupuncture and moxibustion on the expression of total protein and the expression of BDNF protein in hippocampus,colonic tissue and the expression of BDNF protein in the treatment of functional diarrhea,The synergistic mechanism of the disease,acupuncture for the treatment of the clinical selection of the disease and provide a scientific basis.Methods:Fifty SPF grade SD rats were randomly divided into blank control group,model group,electroacupuncture group,electroacupuncture group and electroacupuncture group.In addition to the blank group,the other four groups of rats were senescent gavage method to copy functional diarrhea model,the blank group was treated with the same dose of saline.Electroacupuncture at acupoints,electroacupuncture at the acupoint group,electroacupuncture at the acupuncture points group 1h after daily administration of electroacupuncture treatment,acupuncture time 15 min,once a day,a total of 10 days.At the end of acupuncture treatment,10 rats in each group were anesthetized,and the colonic tissue was taken from the abdominal cavity.The hippocampus was taken from the craniotomy and the colony tissue and hippocampal tissue protein concentration were measured by total protein assay.At the same time,the expression of BDNF protein in colonic tissue and hippocampus was detected by biotin double antibody sandwich enzyme-linked immunosorbent assay.Results:1.Compared with the model group,the water content of the feces in the model group was significantly higher than that in the control group(P <0.01).Compared with the model group,the water content of the 24 h manure in Shu group and Shu-Mu group were decreased(P <0.01).2.Compared with the blank group,the number of defecation in the model group increased(P <0.01).Compared with the model group,the number of defecation particles decreased after 1 hour of intragastric administration in Mu group(P <0.01).3.There was no significant difference in the content of BDNF protein between the two groups(P> 0.05).4.Compared with the blank group,the expression of BDNF in hippocampus of model group increased(P <0.05).Compared with the model group,the hippocampus BDNF in Shu group and Shu-Mu group increased(P <0.05).5.There was a significant negative correlation between BDNF protein content in colon and hippocampus of Shu group(P <0.05).6.Compared with the blank group,the colonic protein concentration in the model group was significantly decreased(P <0.05).Compared with the model group,the colonic protein concentration in Mu group and Shu group increased(P <0.05).7.Compared with the blank group,the total protein concentration of the hippocampus was significantly decreased in the model group(P <0.05).Compared with the model group,the concentration of hippocampal protein in Mu group and Shu group increased(P <0.05).8.There was no significant correlation between colonic and hippocampal protein concentrations in each group(P> 0.05).Conclusion:1.Electroacupuncture can relieve the symptoms of diarrhea in FD rats,Shu-Mu acpoints exercise the spleen and stomach work.2.Shu group can increase the level of hippocampus BDNF,causing colonic,hippocampal tissue protein content changes;Mu group can increase the hippocampus protein changes;Shu-Mu group can be improved by increasing the level of hippocampal BDNF and colonic protein in the treatment of FD rats.
