Study On The Regulatory Mechanism Of Periplaneta Americana Extract On Cell Apoptosis In Chronic Refractory Wounds

objective To observe how the American extraction solution regulate apoptosis genes by the bcl-2 ? bax and caspase3 in the chronic wound healing and explore its healing mechanism to the chronic wound.Methods The60 SPF SD lab rats were divided randomly into the acute wound control group,the chronic wound groups including the model group,the American extraction solution experimental group and the Beifuji positive group averagely(n=15).After the rats models building successfully,the research group changed dressings by the corresponding medical reagents and kept a part of wound tissue after anesthesia to rats in batches on 3nd,8th,15 th,20th day and executed the whole rates finally.Expression quantity of the bcl-2?bax and caspase3 in wound tissue was measured by Realtime polymerase chain reaction(RT-PCR).Results The expression comparison between the model group and the other groups in bax,bcl-2,caspase3 is significant statistically(P < 0.05).The result of express comparison in bax,caspase3 mRNA between groups are almost uniformly.From day 3 to day 20,the expression quantity of the model group is higher than the experimental group and positive group.the control group changes slowly and its expression quantity are the lowest among the four groups(P < 0.01),The comparision between the experimental group and the positive group is not obvious(P > 0.05).The result of bcl-2 is contrary,the expression quantity of the bcl-2 of the control group is higher than the model group,the positive group and the experimental group(P <0.01).On the 3rd day,the expression quantity of the experimental group is lower than the positive group and it is on the rise from the 8th to the 15 th day and it declines slightly on the 20 th day.the expression quantity of the experimental group is higher than the positive from the 8th to the 20 th day.Conclusion The American extraction solution can regulate the expression of bax/bcl-2 and caspase3 RNA in chronic wound healing effectively and optimize the mechanism of the cell apoptosis that promoting the chronic wound repairment and reducingthe scar formation.
