Objective: To observe the rat model of LDH successfully modeled by replantation of autologous nucleus pulposus to investigate the effect of Bugan Jianyao Decoction on expression of NF-?B m RNA and IL-1 in rat lumbar disc herniation(LDH)tissue and discus the possible mechanism of Bugan Jianyao Decoction treating LDH.Methods: One hundred SD rats were randomized into the normal control group(n = 20)and model group(n = 80).Among the model group,60 were modeled by replantation of autologous nucleus pulposus and successfully modeled rats with LDH were randomly assigned to the control group,Bugan Jianyao Decoction group and celecoxib group,and the rest of the 20 ones to the sham group.For sham group,rat vertebral laminae were removed and L5 nerve roots were merely exposed merely,and replantation of autologous nucleus pulposus was not given.One week after successful modeling,both Bugan Jianyao Decoction group and celecoxib group were intervened with the corresponding drugs,while the normal control group,the sham group and the model group were given the same amount of normal saline.During the course of the experiment,animal behaviors were observed and neurological function was scored for model rats.All groups of animals were sacrificed in batches for their embedded disc tissues and surrounding tissues before,7,14,and 21 days after drug intervention.HE-stained specimens were observed histomorphologically for embedded disc tissues and surrounding tissues under a light microscope(400 ×).Content of NF-?B m RNA and IL-1 in embedded disc tissues and surrounding tissues was assayed by RT-PCR and IHC,respectively.Results: Comparison of normal control group,model group,Bugan Jianyao Decoction group and celecoxib group after modeling found abnormal animal behaviors and declined neurological function.Microscopical findings showed histomorphological changes,and significantly high levels of NF-?B m RNA and IL-1were detected.In the sham group,histomorphological changes and low expression of NF-?B m RNA and IL-1 were found.After drug intervention,animal behaviors and neurological function became more normal,histomorphological changes improved,and expression of NF-?B m RNA and IL-1 was down-regulated for the Bugan Jianyao Decoction group and the celecoxib group;for the normal control group and the sham group,there was no significant change before and after intervention;for the model group,animal behaviors and neurological function were abnormal,expression of NF-?B m RNA and IL-1 was up-regulated further.Conclusion: 1.Replantation of autologous nucleus pulposus can successfully establish a rat model of LDH,which can detect high levels of expression of NF-?B m RNA and IL-1,indicating that positive expression of NF-?B m RNA and IL-1 in the body is one of the pathogeneses of LDH.2.Both Bugan Jianyao Decoction and celecoxib can down-regulate the expression of NF-?B m RNA and IL-1 in the embedded nucleus pulposus of the rat model of LDH,and there is no significant difference in degree of intervention of this index between Bugan Jianyao Decoction and celecoxib capsules.3.Effect of Bugan Jianyao Decoction on levels of expression of NF-?B m RNA and IL-1 is related to the duration of drug intervention,i.e.,longer duration of drug intervention leads to more significant down-regulation of expression of NF-?B m RNA and IL-1.
