The Effect Of Bugan Jianyao Recipe On The Expression Of IKK?mRNA And TNF-? In Rat Lumbar Disc Herniation Tissue

Objective: To investigate the effect of Bugan Jianyao Decoction on expression of IKK-?m RNA and TNF-? in rat lumbar disc herniation(LDH)tissue.Methods: According to the random number table,20 of 100 SD rats were randomly assigned to the normal control group.Of the rest of the 80 rats,20 were randomly assigned to the sham group,60 were modeled into rat models of LDH(L5/L6).Their behavioral and morphological changes were observed and neurological function was scored for each group of rats.Successfully modeled rats were then assigned to the model group(n = 20),Bugan Jianyao Decoction group(n = 20)and celecoxib group(n = 20).All rats were equally assigned to five groups in total.Normal control group received no treatment and normal saline was administered by gavage;for sham group,L5 nerve roots were merely exposed,and autogenous caudal disc compression was not given but normal saline was administered by gavage;for model group,Bugan Jianyao Decoction group and celecoxib group,normal saline,Bugan Jianyao Decoction,and celecoxib were administered by gavage,respectively.Caudal disc tissues embedded at L5 nerve roots by autograft were sampled at 1,2,and weeks after postoperative medication.HE-stained specimens were observed histomorphologically for caudal disc tissues under a light microscope(400 ×).Expression of IKK-?m RNA and TNF-?in rat LDH tissue was assayed by RT-PCR and IHC,respectively.Results: 1.Microscopically,significant morphological changes of different degrees were observed in modeled rat LDH tissues before,1 and 2 weeks after drug intervention;for Bugan Jianyao Decoction group and celecoxib group,microscopically,rat LDH tissues were repaired and improved 3 weeks after drug intervention compared those before intervention,and the difference was not significant between both groups.2.Assayed and found after modeling,expression of IKK-?m RNA and TNF-? in rat LDH tissue was up-regulated significantly,after intervention with Bugan Jianyao Decoction and celecoxib,expression of IKK-?m RNA and TNF-? was down-regulated significantly,and there was little difference between Bugan Jianyao Decoction group and celecoxib group.Conclusion: 1.Rat model of LDH can be successfully established by autogenous caudal disc graft and embedding and it enables up-regulation of expression of IKK-?m RNA and TNF-?.2.IKK-?m RNA and TNF-? are upstream and downstream mediators of NF-?B signaling pathway,respectively,while Bugan Jianyao Decoction can cause down-regulation of expression of both,suggesting that the pathogenesis of LDH is closely related to the NF-?B signaling pathway.3.Both Bugan Jianyao Decoction and celecoxib can down-regulate the expression of IKK-?m RNA and TNF-? in a rat model of LDH,which have an intervention effect on inflammatory response to LDH,without difference between them.Traditional Chinese medicine may be one of the mechanisms underlying the treatment of LDH.
