| Object:To observe the effect of Bu Gan Jian Yao Fang on the expression of MMP-1 and OP-1in the nucleus pulposus of rats with lumbar disc degeneration,and to explore its therapeutic effect and mechanism on the treatment of lumbar disc degeneration from the cellular biological level.Methods:34 of 66 3-4-month-old clean healthy SD male rats were randomly selected to establish the model by the whole layer puncture of lumbar fiber ring.After the model was made successfully,the rats were divided into two groups by the random number table method: the group of Bu Gan Jian Yao Fang,the group of model,12 rats in each group,were fed with Bu Gan Jian Yao Fang(containing crude drug 2.412g)and normal saline respectively.The remaining 24 rats,12 in the normal control group and 12 in the sham operation group.The normal control group was not treated,the sham operation group only exposed the intervertebral disc,did not carry out lumbar disc puncture,all were fed with normal saline.Four rats in each group were killed at the time points of 2,4 and 6 weeks after administration respectively.The degenerative segments of L4/ 5 and L5/6 nucleus pulposus of intervertebral disc were taken.After HE staining,the histomorphology of the samples was observed,the content of MMP-1 in L4/5 disc nucleus pulposus was detected by RT-PCR,and the content of OP-1 in L5/6 disc nucleus pulposus was detected by ELISA.Results:1.Microscopic observation of lumbar intervertebral disc tissue in rats after modeling showed changes of nucleus pulposus cell reduction and annulus fibroproliferation before drug intervention,the damaged structure of the nucleus pulposus of the lumbar intervertebral disc tissue of the model rats was improved obviously after the drug intervention of Bu Gan Jian Yao Fang.2.After building of MMP-1 in rat lumbar disc tissue expression increased,the content of the OP-1 expression from the previous fall,use for liver health waist party drugs after the intervention of MMP-1 in rat lumbar disc tissue expression appears to reduce,the content of the OP-1 expression from the previous rise.Conclusion:1.The degenerative model of lumbar intervertebral disc can be successfully reproduced by the whole layer puncture of lumbar fiber ring.2.In the experiment,MMP-1 and OP-1 in the model group were changed compared with the normal control group and the sham operation group,which proved that the lumbar disc degeneration was related to the expression of MMP-1 and OP-1.3.Bu Gan Jian Yao Fang can down regulate the expression of MMP-1 m RNA and promote the expression of OP-1,which may be the mechanism of its treatment of lumbar disc degeneration. |
