Objective To develop rat model study of experimental lumbar degenerative interevertebral disc and detect the expression of bFGF and MMP-13 in the disc nucleus pulpous tissue from normal disc,degenerative disc and degenerative disc treated by celecoxib using immumohistochemistry and Western-Blot to explore its value in lumbar degeneration disc and treatment with celecoxib inhibits the expression of bFGF and MMP-13 in the rat lumbar degenerative disc .Methods 96 SD rats were randomly divided into four groups: untreated control group(Group A),degenerative control group((Group B), treatment group of small dose of celecoxib (Group C) and treatment group of big dose of celecoxib (Group D),Every group had 24 rarts.The rats were operated,exscinding the supraspinal and interspinal ligament ,half of the upper and lower lumbar facet joints and bilateral erector muscle between the first and the sixth lumbar sipoine. After 2 weeks from operation ,the rats of group C and group D were underwent gavage with celecoxib among 10mg/kg /day and 25mg/kg /day for 2 weeks. 1,2,3month after operation respectively,4 rats were chosen randomly and respectively from every group . Lumbar discs tissues were obtain ,the expression of bFGF and MMP-13 were examined by the method of immunohistochemistry and Western-Blot .Results The results of pathologic examination indicated that the nucleus pulposus cells unevently distributed throughout the disc. the number of nucleus pulposus cells decreased. Under immumohistochemistry, there was significant difference in IOD value among treatment group of celecoxib and control group(P <0.05).There was no significant difference in IOD value among treatment group of celecoxib and untreated control group.The expression of bFGF and MMP-13 in treatment of celecoxib were much less than those in degenerative control group by Western-Blot(P <0.05). Co-expression of the bFGF and MMP-13 exist in degenerative disc tissues.The protein content of them in degenerative disc has a significant correlation ( P < 0.05).Conclusions The expresstion of bFGF and MMP-13 in lumbar degenerative disc was highly and they were similarily. It indicated that bFGF and MMP-13 may be involved in development of lumbar disc degeneration .Celecoxib as COX-2 inhibitor can downregulate the expression of bFGF and MMP-13 in the disc nucleus pulposus.It will provide the theory evidence for delaying lumbar degenerative disc development.
