Objective Insomnia is one of the common clinical syndromes in medicine.Although not part of critical illness,it can often induce or aggravate heart palpitations,chest pain,dizziness,stroke and other medical disorders.Intractable insomnia not only reduces the work and quality of life,but also induce patients with serious psychological burden and emotional distress.“Shuangxia Soup”prescription was collected in "Chongding Linglan Yaolan"-one of the early medicine handbook.Spoken from the theoretical foundation of Chinese medicine treatment,"Shuangxia Soup" was in line with the rise and fall of the laws of nature,and suits with the rhythm of the human body rest.There has been accumulated a large number of experience in the clinical practice.This study aimed to explore the changes of different compatibility proportion“Shuangxia Soup" in chemical composition and fingerprints.Moreover,it was used by the pharmacodynamic screening model for finding the best compatibility proportion and the active chemical components in“Shuangxia Soup".Methods 1.HPLC-UV method was used to determine the content of trigonelline,uracil and adenosine in different compatibility proportion of“Shuangxia Soup".2.HPLC-UV method was used for the determination of different compatibility proportion“Shuangxia Soup”liquid chromatogram.It was assigned to judge the common components peaks from the chromatogram and compared the differences between them.3.DAMS activity monitoring system was used to determinate the sleep time of control group and treatment group.By the results,we can screen the best delivery time and compare the different compatibility of efficacy differences.4.Using HPLC-ECD method to determine the 5-HT,DA and NE,HVA content in drosophila brain.According to changes of various monoamine neurotransmitters content,we can judge the treatment efficacy and screen the best compatibility proportion"Shuangxia Soup".Results1 The compatibility between Pinellia and Prunella can promote the dissolution of uracil and adenosine,but the opposite of trigonelline.2 It was used by HPLC to measure liquid chromatograms of the different compatibility proportion of Shuangxia Soup.From it,we got 12 chemical component peaks.Compared with the original crude drug liquid chromatograms,we could conclude that peaks 3,4,10,11 belongs to pinelli,peaks 8,9 belongs to Prunella,the rest is common belongs to both.Comparing the areas of the peak 3,4,8,9,10,11,it can be found that with the increase of prunella,the content of these ingredient was gradually increase.3 Drosophila's sleep time was monitored by the system DAMS to study the effects of different compatibility proportion of "Shuangxia Soup" on the sleep behavior of Drosophila.The results showed that the optimum drug incubation time is 3 days,the best compatibility proportion of Shuangxia Soup was 1:1 for mostly prolonged sleep time in drosophila.4 The drosophila were grouped by the control group,sleep deprivation group,the treatment group and sleep deprivation treatment group.It was used by HPLC-ECD to measure 5-HT,DA,NE and HVA levels in Drosophila head.By comparing different neurotransmitter levels,the optimum ratio for Shuangxia Soup for promoting sleep of drosophila was 1:1.Conclusion Considering the adenosine,uracil,trigonelline role in the treatment of sleep,the compibility decoction between pinellia and prunella showed better results than the single.Seen from comparison among the different proportions of Shuangxia Soup liquid chromatogram,some ingredients increases with the increase proportion of prunella in the compatibility.Since prunella was rarely reported efficacy in the treatment of insomnia,it can be inferred the existence of the maximum dose and the optimal dose for prunella in the Shuangxia Soup.Drosophila activity monitoring of sleep and head monoamine neurotransmitters measured results show that the optimal proportion for promoting drosophila sleep was 1:1.The experiment failed to find the chemical composition(s)which can show significant difference reflected the optimal ratio.This question would be the focus of latter study.Since the experiment was studied from two levels-basic chemicals and pharmacodynamics screening-to verify that the compatibility between pinellia and prunella 1:1 is reasonable-which is consistent with clinical applications,it can also provide the guidance reference.for the study of a similar material basis and mechanism of Chinese herbal compound.
