| In this research, an anti-cancer drug screening model based on apoptosis-induction was established using Drosophila melanogaster The effects of media on the development of Drosophila were studied. Two strains of Drosophila, the strain containing FLP/FRT recombination system marked with green fluorescence protein (GFP) (T1) and the scrib/TM6B balance strain (T2) were used for hybridization, leading to a tumor model that has GFP-scrib-/- mosaic in the imaginal disc of eyes.Observed with an optical microscope, the adults of the first filial generation (F1) had abnormal hyperplasy in eye tissues, which displayed strong green fluorescence under a fluorescence microscope (excited at 450-490 nm). In the patterns of protein SDS-PAGE and Western blot, the samples of T1×T2 contained a band at about 20 kD, which was absent in the other samples. It is indicated that the expression of GFP and abnormal hyperplasy in the eye tissue are linked. In addition, we are trying to use fluorescent microplate reader to quatitatively test this link,but it failed. The compounds of synthetic drugs and JNK agonis were added in the modified medium quatitatively during culturing.The preliminary results didn't show obvious effects on the development of Drosophila and tumor forming. |
