Insomnia is a very common symptom with high economic and personal costs associated with decreased quality of life and functioning.It is a common sleep disorder characterized by difficulty falling asleep,staying awake,or both.Insomnia is common all over the world either.At present,a proportion of insomniac patients worldwide resort to many kinds of complementary and alternative medicine(CAM).Traditional Chinese medicine(TCM)including Chinese herbal medicine has played a significant role in the medical care of insomnia patients for more than 2000 years.Shuangxia Decoction(SXD)is a prescription founded by Wang Kentang,a famous doctor in Ming dynasty.SXD is composed of two medicinal herbs:Rhizoma Pinelliae Praeparatum and prunella vulgaris.Pinellia ternata(Thunb.).In the traditional theory of traditional Chinese medicine,Pinellia belongs to Yin and Purnell belongs to Yang.In common function restores the balance of Yin-Yang of body energy so the body's normal function.Based on the preliminary study of the material basis,the mechanism of the Shuangxia Decoction was described.Involves the main content includes:1.Pharmacodynamics study on curing insomnia by Shuangxia Decoction regarding Drosophila melanogaster as the model organismTo explore the influence of freeze-dried powder from water decoction of Shuangxia Decoction(SXD)to the sleep condition of normal fruit flies and fruit flies whose sleep is divested by light.Female fruit flies with age of 7 days had similar sleep activity with human beings of doing activity in day time and sleeping at night.There is no significant difference in the sleep time of 3-7 days virgin flies,and it is relatively stable.Plateau appears after the concentration of 2.5%.The total sleep time does not increase obviously and shows downtrend after this concentration.The sleep time of fourth day is the longest time after giving SXD.After giving high,middle and low concentration decoction with SXD to fruit flies whose sleep is divested,it shows middle dosage group(with medicine concentration of 2.5%)has the best insomnia curing effect.Positive medicine group has obvious difference with group whose sleep is divested.Freeze-dried powders from water decoction of SXD effectively improve sleep of fruit flies.Middle dosage group has obvious curative effect to fruit flies who are insomnic stimulated by light.The software can be used in traditional Chinese medicine(TCM)pharmacodynamics research.2.Study on the content of monoamine transmitters in Drosophila head by HPLC-ECDThe mobile phase was 50mmol/L sodium acetate(pH 3.4)and methanol.EDC detection voltage was+0.75V,C18 Atlantis chromatographic column(2.1 x 150mm,3 m)was used.The flow rate was kept constant at 0.25mL/min.The ratio of methanol to buffer salt solution was 4:96.The method was validated thoroughly in this thesis,and the result exhibited enhanced specificity,accuracy and repeatability This method was aslo easy to operate,convenience to achieve,and suited for routine monitoring of 5-HT?DA?NE?5-HIAA?HVA in biological fluids.3.Study on mechanism of SXD treating insomnia disease through network pharmacology.With the rapid progress of bioinformatics and systems biology,network pharmacology is considered as a promising approach toward reveal the underlying complex relationship between an herb and the disease.In order to discover the mechanism of SXD treating insomnia disease systematically,we use the auxiliary mechanism elucidation system for Chinese medicine,built up a molecule interaction network on the active component targets of SXD and the therapeutic targets of insomnia disease to offer an opportunity for deep understanding the mechanism of SXD treating insomnia disease from the perspective of network pharmacology.The results showed that SXD treating insomnia disease through some pathways as follows:G-protein coupled receptor protein signaling pathway,second-messenger-mediated signaling,cellular calcium ion homeostasis,synaptic transmission,regulation of cAMP biosynthetic process,neuropeptide signaling pathway.
