"Herbs is good,medicine is good",good internal quality properties,only after the effective delivery of preparation technology,medicine can be good.This research group proposed the theory of component structure,and the research results showed that the reason for the superior quality and effect of authentic herbs was not the inclusion of a particular component,nor the higher the content of a certain component,but the volume-ratio relationship between ingredients/components is different from that of non-authentic Chinese crude drugs,with the unique structural characteristics of the material base components.However,it is worth further studying whether the structural characteristics of the components of crude drugs are reflected in the preparation products and the changes in each process after the corresponding preparation process.Therefore,this study further discussed the structural characteristics of the components of genuine Moutan Cortex's superior potency,and analyzed the structural characteristics of the components of the main active substances derived from Moutan Cortex in Liuwei Dihuang concentrated pills,so as to explore the transfer rules in the process.This study provides a scientific basis for the transfer analysis of genuine component structure of Moutan Cortex in the preparation process of Liuwei Dihuang concentrated pills.The main contents include the following parts:1.Study on the structural characteristics of the material base?secondary metabolites?of genuine Moutan CortexThe essential difference between traditional Chinese medicine and non-traditional Chinese medicine is the difference in the structural proportion of the underlying ingredients/components.In order to reveal the structural characteristics of genuine Moutan Cortex,20 batches of genuine Moutan Cortex and 12 batches of non-genuine Moutan Cortex were collected and analyzed.HPLC fingerprint and similarity analysis were used to study the inner quality difference of Moutan Cortex from different producing areas.By means of Clustering Analysis?CA?,Principal Component Analysis?PCA?and orthogonal partial least squares discriminant analysis?OPLS-DA?,the structural characteristics of a material base component are extracted.HPLC method was used to determine the content and volume ratio of representative ingredients/components of genuine Moutan Cortex and non-genuine Moutan Cortex.The results show that there is a certain difference in the inner quality of genuine Moutan Cortex and non-genuine Moutan Cortex,and genuine Moutan Cortex has the unique structural characteristics.It was found that the quantitative and quantitative ratios of terpene glycosides,tannins and phenolic acids were 14.1 mg·g-1,12.5 mg·g-1,21.7 mg·g-1,1.00:0.89:1.54.In samples S28 and S32,the ratio of component to component was more similar to that of genuine ground Moutan Cortex.However,the amount of three components in S28 of genuine ground Moutan Cortex produced in Hebei province was significantly different from that of genuine ground Moutan Cortex,and the amount of S32 component of Moutan Cortex from Zhongjing Wanxi planting base in Bozhou of Anhui province was similar to that of genuine Moutan Cortex.The results showed that the quality of Moutan Cortex sample S32 was closer to that of genuine Moutan Cortex.2.The component characteristics of inorganic elements in genuine Moutan Cortex and its difference with other areas were studiedIn recent years,most of the researches on the genuine properties of Moutan Cortex concentrate on the secondary metabolites,but few on the inorganic elements.Therefore,It is very important for the identification and safety evaluation of genuine and non-genuine Moutan Cortex to comprehensively analyze and master the content and distribution of inorganic elements.In this chapter,based on the theory of"component structure",the inorganic elements in genuine and non-genuine Moutan Cortex were analyzed by inductively coupled plasma mass spectrometry?ICP-MS?,and the fingerprints of inorganic elements were drawn.Through correlation analysis and principal component analysis,the constituent elements of inorganic elements in Moutan Cortex?characteristic elements?were excavated.The quantitative and volume-ratio relationships between the components of genuine and non-genuine Moutan Cortex samples were clarified.The results show that the genuine Moutan Cortex has a unique structure of inorganic elements which is different from other non-genuine Moutan Cortex,and the amount of characteristic elements in the original series contained genuine Moutan Cortex was 0.33 mg·g-1,1.42 mg·g-1,1.33 mg·g-1,0.11 mg·g-1,4385.78 mg·g-1,4.87 mg·g-1,0.49 mg·g-1,respectively,volume ratio relationship for?1.00:4.30:4.03:0.33:13290.24:14.76:1.48?,the amount of characteristic elements in the original packing genuine Moutan Cortex was 0.24 mg·g-1,1.96 mg·g-1,0.36 mg·g-1,0.05 mg·g-1,5122.01 mg·g-1,4.81 mg·g-1,0.05 mg·g-1,respectively,volume ratio relationship for?1.00:8.17:1.50:0.21:21341.71:20.04:0.21?.3.The structural characteristics of the components of Moutan Cortex were analyzed on the differences of the transfer rules in Liuwei Dihuang concentrated pills from several manufacturersOne of the raw materials of Liuwei Dihuang concentrated pills was Moutan Cortex,which was prepared into Liuwei Dihuang concentrated pills through related processes.Then,can the unique structural characteristics of components in genuine Moutan Cortex be transferred to Liuwei Dihuang concentrated pills?This chapter analyzes the structural characteristics of the main active material components in Liuwei Dihuang concentrated pills,which originated from Moutan Cortex,in order to discuss the transfer rules in the process.HPLC was used to characterize the components of Moutan Cortex in Liuwei Dihuang concentrated pills.The differences of the transfer rules of the components of Moutan Cortex in Liuwei Dihuang concentrated pills from different manufacturers were clarified.The results showed that the components,content and volume ratio of Moutan Cortex varied greatly in the concentrated pills of Liuwei Dihuang from several manufacturers.The ratio of terpene glycosides,tannins and phenolic acids in LwDh21-LwDh29 of Liuwei Dihuang concentrated pills fluctuated little,and the component structure was basically stable.In addition,there was a certain similarity in the volume ratio relationship among oxidized paeoniflorin,paeoniflorin and benzoyl paeoniflorin in terpene glycosides components of the samples LwDh1,LwDh2,LwDh6,LwDh10,LwDh11,LwDh16,and LwDh21-LwDh29.And there was a significant difference in the volume ratio relationship among the groups of other batches of preparation products.4.ConclusionBased on the theory of"component structure",this paper further discusses the structural characteristics of the optimum potency of genuine Moutan Cortex.The structural characteristics of the main active material components in Liuwei Dihuang concentrated pills derived from Moutan Cortex were analyzed to discuss the transfer rules in the process.It provides a scientific basis for the transfer analysis of genuine components of Moutan cortex in the preparation process of Liuwei Dihuang concentrated pills.
