| Tricholoma matsutake,as a type of precious and rare edible fungus,contains abundant bioactivity nutrition has been widely noted for its healthy function and medicinal value.But for the restriction of season,geography,climate and other factors,it is difficult to fresh-keeping,a large number of bioactivity component was lost.Matsutake fresh-keep became a worldwide problem.How to preserve matsutake nutrients and make a long-term storage are problems badly needed to be solved and a object for future research.My experiment got "liquid matsutake" contains full nutrients and longer storage stage.Its healthy funtion was verified by mice experiments.The main conclusion are as follows:(1)Optimized ultrasonic extraction with water:125 w for 10 min,ultrasonic working time 4 s,interval 1 s;optimized weak ionic extraction:sodium salt solution concentration of 0.3%saline concentration of 0.5%,extraction time 120 min,extraction temperature 80℃;optimized solution extraction:80%ethanol,extraction time 30 min,extraction temperature 70℃.Under optimum conditions the measured ultrasonic assisted water-soluble polysaccharide extracted mushroom yield 0.52 g/100g,vitamin C yield 3.0 mg/100g,flavonoids yield 7.1mg/100g;weak base yields an alkaline salt solution extracted proteins reach 1.16 g/100g;alcohol solution and extracted polyphenols yield triterpenoids also reached 1.16mg/100g and 0.23 g/100g.(2)Concentrate on the quality of oral matsutake made a preliminary assessment of their senses to detect,relative density,pH,hygiene indicators and nutrient content.The results show that the clarity of the mushroom was concentrated oral brown liquid,no turbidity and precipitation,sweet,relative density was 1.10±0.05 g/mL,pH value of 7.50±0.05,the amount of each device 10 mL.brown bottles,hygiene testing standard,100 mL containing polysaccharide 86.7±0.2 mg,vitamin C 0.50±0.04 mg,flavonoids 1.18±0.04 mg,protein 193.3±0.3 mg,polyphenol 0.19±0.02 mg,triterpenoids 38.3±0.1 mg,detected in 17 kinds of amino acids.its good quality.(3)In this experiment,mice immunity,antioxidant activity,anti-fatigue function as a check indicators to mice fed a normal saline as a control to compare the changes matsutake concentrated oral gavage in mice.The results showed that,in immunoassays,compared with the normal saline group,with the increase in the number of days gavage,thymus index,spleen index,serum IgG and macrophage phagocytosis in continuous improvement,when administered 21 d thymus index improved by 26%,spleen index increased by 9.1%,serum IgG levels increased by 11%to improve macrophage phagocytic capacity by 51%,the difference was significant(P<0.05);antioxidant test,and normal saline group phase compared with the increase of the number of days gavage,MDA concentration declining,SOD activity is rising,when administered 21 d MDA content decreased by 57%,SOD content has increased by 11%,the difference was significant(P<0.05);fatigue detection,compared with the normal saline group,with the increase in the number of days gavage,swimming time improved by 39%,and blood urea nitrogen and serum lactate levels were reduced by 29%and 49%.In conclusion,oral liquid with multi-step extraction had high nutritional value,and good quality.Animal tests showed matsutake can improve immunity,antioxidant and anti-fatigue. |
