Tricholoma matsutake was a kind of edible and medical fungus. Polysaccharides from Tricholoma matsutake possess a wide variety of physiological activities, such as antitumour, anti-aging, immunomodulating, etc. The submerged fermentation process, extraction, physical and chemical properties, immunological function of polysaccharides from Tricholoma matsutake were studied.By orthogonal test, the results showed the optimized fermentation medium was glu-cose4%, peptone soy4%, yeast extract0.2%, MgSO4-7H2O0.15%, KH2PO40.3%.Meanwhile the effects of pH, medium capacity, inoculation quantity, culture temperature, shaking frequency on the mycelia growth and exopolysaccharides were studied. The optimum shaking culture condition was as follow:initial medium pH6.0,10%inoculation quantity,25℃,170r/min shaking frequency,100mL medium in250mL shaking flask,6days incubation. The ferment condition of10L fermentor was:25±℃,200r/min spurring speed,3days incubation. Productivity of39.1g/L was obtained.The suitable procedure for extraction exopolysaccharides from Tricholoma matsutake was determined as follow:pH of ferment liquid was changed to6.0, concentrated to1/2volume at60℃and then precipitated at a final75%ethanol at4℃for24hours. The precipitate was harvested by centrifuge, washed several times with100%ethanol.The suitable procedure for the intracellular polysaccharide from Tricholoma matsutake was determined as follow:the ultrasonic power was400W for30minutes, the extracting time was1.5hours at90℃, and the ration of solid to liquid was1:30.The animal experiments demonstrated that the exoploysaccharides and intracellular polysaccharide (containing protein) could enhance macrophage phagocytosis and lymphocyte transformation. The exoploysaccharides (containing protein) and exoploysaccharides (not containing protein) on the antibodies generated had a very significant role.
