Biodiversity investigations in specific areas is an important basic work for the conservation of biodiversity,the evaluation of species survival status and the rational exploitation and utilization of species resources.As one of the important components of biodiversity,macro-ascomycetes resources lack the investigations of diversity in specific regions.This study conducted a field survey and assessment of macro-ascomycetes resources in Xingshan County,Hubei Province,and conducted molecular systematic studies based on the obtained genetic data to initially explore its evolutionary relationships.All the specimens were collected in more than 20 sites in 6 towns of Xingshan County,spanning the spring,summer and autumn,covering a variety of forest types.A total of 416 specimens were obtained from the collection,belonging to 4 classes,5 orders,21 families and 51 genera.Among them,339 specimens were identified as species,belonging to 4 classes,5 orders,20 families,48 genera and 96 species,77 specimens were identified as genus.And the species list of macro-ascomycetes in Xingshan County was compiled.Among them,there are 40 new records in Hubei Province;10 edible fungi,10 medicinal fungi,4 edible and medicinal fungi,3 toxic fungi;44 geophilous fungi,43 lignicolous fungi,and 10 entomogenous fungi.Species composition analysis found that the genus having the largest number species were Peziza,indicating that the genus is a dominant genus in the region.The geographical distribution of macro-ascomycetes in Xingshan County found that the macro-ascomycetes in Xingshan County were concentrated in the area of 800-1600 m above sea level with typical subtropical evergreen deciduous broad-leaved mixed forest.The most representative areas are Longmenhe Village,Wanchao Mountain,Zhenzi Township.This study obtained a lot of rDNA-ITS and LSU sequences.Combining the reliable sequence in the NCBI's database and the obtained self-measured data,the life tree of macro-ascomycetes in Xingshan County was constructed using Bayesian method and Maximum Likelihood method.And the dominant genus Paziza in Xingshan County were respectively analyzed for phylogenetic analysis using Bayesian method,Maximum Likelihood method,Maximum Parsimony method;the main species Peziza succosa,were analyzed for genetic diversity.Molecular phylogenetic studies on the life tree of macro-ascomycetes in Xingshan County supported the classification treatments of some units of Ascomycetes;the phylogenetic analysis of the genus Peziza found that the species of the genus Peziza in Xingshan County is rich.The study on the genetic diversity of Peziza succosa found that the gene flow was frequent between the geographical units of Xingshan County.The results of this study supplemented the information of macro-ascomycetes resources in Hubei Province,enriched the information of genetic data,fully recognized the diversity of macro-ascomycetes in Xingshan County,and was beneficial to the scientific protection and rational application of macro-ascomycetes in Xingshan County.
