Microfluidic chip is a microdevice that is capable of accomplishing chemical or biology functions through manipulating nanoliter or picoliter fluids or droplets in micrometer-scale structures.It has many advantages including less consumption of samples and reagents,rapid mixing,without cross contamination and good repeatility.Due to the growth environment of the cell can be accurately controlled to make it not or hardly affected by the external environment,microfluidic chip technology culture research is widely applied to the cell,such as cell co-culture,3D culture.In recent years,the establishment of hypoxic environment in the microfluidic chip and the biological behavior of exploration target cells in a hypoxic environment are more report.This paper reports a microfluidic device that can realize hypoxic environment.Different from the common microfluidic chip,through the design of chip structure and the introduction of chemical reaction,the constructed microfluidic device not only has the advantages of simple operation,and oxygen concentration on the formation of hypoxic environment is not single,but a concentration gradient,and the influence of the chip structure,concentration of reactant and flow rate on the oxygen gradient distribution are explored.Hypoxia oxygen concentration gradient of the microfluidic device are applied in myocardial cells culture,to explore the effect of hypoxia on cell growth,but also due to the oxygen gradient exists,can compare more directly caused by the different oxygen concentrations,and explore the influence of hypoxia on the growth of myocardial cells.
