Nowadays,the concentration gradient chips are widely used in many fields,such as microbiological detection,drug screening,toxicological experiments and many other fields,it has great significance in rapid detect the optimal inactivation concentration of bacterial viruses.Compared with the traditional artificial plate configuration with different concentrations of reagents,using the concentration gradient generator will save many complicated operation,the configuration of sample is more accurate and quick.So,it has particularly advantages.In this paper,a large flux density gradient generator and an automatic liquid adding system based on microfluidic are proposed.The system can automatically generate specific concentration gradients and automatically add the samples to the orifice plate by FPGA control.From the preceding discussion the channel analogy on the microfluidic chip is the circuit,the liquid in channel analogy is current.Using the Kirchhoff theorem in circuit principle to design the microfluidic channel.Drawing chip structure by drawing software AutoCAD.Simulation software Ansys Fluent is used to simulate the concentration gradient generator chip.Microfluidic chips are made by photolithography.In the follow-up experiments,the chips are optimized and verified,so that the chip can be used at large flux.The influence of channel length and liquid velocity on the formation of concentration was studied.In the later stage,the accuracy of concentration was observed by fluorescein sodium,and fluorescence intensity was used to characterize the concentration to verifying the accuracy of generating concentration.
