ObjectiveThe changes of body weight,Lee’s index,RBP4 mRNA expression in visceral adipose tissue,protein expression and plasma concentration in aged obese rats were measured by exercise combined with turtle oil.To explore the mechanism of insulin resistance and to provide theoretical reference for the prevention and treatment of obesity and related diseases in the elderly.MethodFifty normal male SD rats,aged 3 weeks,were randomly divided into natural growth group(n = 18)and normal diet(n = 18)with a fat content ratio of 6.0%.The model group(n = 32)was fed with 36% / 40% high fat diet in three stages to establish an old obese rat model.Natural growth rats were randomly divided into two groups: control group(n = 6)and elderly exercise group(n = 6).Twenty-four old obese rats were randomly divided into three groups: obesity control group(n =6),obesity exercise group(n = 6),pure supplement group(n = 6)and exercise combined with turtle oil group(n = 6).Exercise intervention was as follows: the slope of running platform was 0 °,the speed was 15 m / min,each group was 15 min,4 groups / time,the rest time was 5 min,and the total exercise time was 60 min.Turtle oil was given orally 1 hour after exercise,supplemented with 0.3-0.4ml of turtle oil once a day,5 times a week for 8 weeks.Body weight was measured and Lee’s index was calculated once a week,RBP4 mRNA expression and protein expression in visceral adipose tissue and plasma RBP4 concentration,blood glucose and insulin concentration and insulin resistance were measured 8 weeks after intervention.Result1 Body mass and Lee’s indexExercise combined with turtle oil could significantly decrease the body weight and Lee’s index of the aged and obese rats(p < 0.01).The exercise combined withsoft-shelled turtle oil can significantly lower the weight and the Lee’s index in the old obese rats(p <0.01).The body weight and Lee’s index of the aged obese rats were slightly decreased with the supplement of turtle oil,but there was no significant difference(P > 0.05).2 RBP4 mRNA expressionExercise slightly increased the expression of RBP4 mRNA in visceral adipose tissue of aged rats,but there was no significant difference(P > 0.05).The exercise can significantly lower the expression of RBP4 mRNA in the visceral adipose tissue of the old obese rats(p <0.01).The expression of RBP4 mRNA was slightly decreased by pure supplementation of turtle oil,but there was no significant difference(P > 0.05).Exercise combined with turtle oil significantly decreased the expression of RBP4mRNA(p < 0.01).3 RBP4 protein expressionThe expression of RBP4 protein in visceral adipose tissue of aged obese rats was significantly increased(p < 0.01).Exercise could increase the expression of RBP4 protein in visceral adipose tissue of aged rats(p < 0.05).The expression of RBP4 protein in visceral adipose tissue of aged obese rats was significantly decreased(P < 0.01),and the expression of RBP4 protein in visceral adipose tissue was significantly decreased by exercise and exercise combined with turtle oil(P <0.01).4 Plasma RBP4 concentrationThe plasma RBP4 concentration in aged obese rats was significantly increased(p < 0.05).The plasma RBP4 concentration in obese rats was significantly increased(p < 0.05).Exercise alone supplemented with turtle oil and exercise combined with turtle oil could decrease the plasma RBP4 concentration(p < 0.01).5 Insulin resistanceBlood glucose and insulin resistance were significantly increased in aged obese rats,but exercise had no significant effect on blood glucose concentration and insulin resistance in aged rats(P > 0.05).Exercise and exercise combined with turtle oil decreased blood glucose concentration(P < 0.05)and insulin resistance(P < 0.05,P< 0.01)in aged obese rats.Supplementation of turtle oil could decrease the bloodglucose concentration(P < 0.05)and insulin resistance(P < 0.01)in the aged obese rats.Conclusion1 The exercise and exercise combined with the soft-shelled turtle oil can reduce the weight of the senile obesity rats and the Lee’s index,and has a better weight-reducing effect.2 The blood glucose and insulin resistance increased significantly in the aged obese rats.The expression of RBP4 mRNA and protein in visceral adipose tissue and the concentration of RBP4 in the plasma of the aged obese rats were significantly higher than those in the aged obese rats.Turtle oil significantly decreased the expression of RBP4 protein.Exercise and exercise combined with turtle oil can significantly reduce the expression of mRNA,protein and plasma RBP4 concentration,and the protein expression has a good synergistic effect.3 Exercise and exercise combined with turtle oil improve insulin resistance and reduce blood sugar.This study suggests that exercise and exercise combined with turtle oil can improve insulin resistance,which may play a physiological role by decreasing the expression of RBP4 mRNA and protein in visceral adipose tissue and the concentration of RBP4 in plasma.
