Objective:Insulin resistance has been become an important problem of human health, the prevention and treatment of cardiovascular disease induced by insulin resistance to be the research emphasis of the medical treatment. Because of the main fatty acid oxidation on normal myocardial energy supply, the role of cardiac muscle in regulating the body glucose homeostasis is not obvious. Therefore, compared with the main organ skeletal muscle, liver, adipose tissue, myocardial insulin resistance in glucose metabolism can not afford a major role in a long time and doesn’t attract enough attention. The harm of myocardial insulin resistance becomes the focus of research in recent years. Myostatin (MSTN) was shown to inhibit muscle growth, inhibition of AKT activity such as insulin resistance. Studies have shown that MSTN as myocardial evil fluid transmission work and studies have shown that the lack of MSTN doesn’t cause the change of heart function and cardiac muscle fiber, the physiological effects of myocardial and skeletal muscle showed obvious differences. This study will get through different ways of movement and the joint motion scheme of MSTN antibody intervention ways on rats model with insulin resistance,which studys from biochemical, morphology, protein, the level of gene different angles.Methods:This study bought healthy male SD rats (Sprague-Dawley) 70 (quality qualified number:shan medical card words 08-004),2 months of age, weight 170~170 g, aseptic environment cage, aseptic environment cage, raising the temperature of 22~25℃, humidity 40~60%, free drinking water, adaptability raised after a week, were randomly divided into normal group (NC,10) and a high-fat diet group (HF,60). Group raised after 3 months, by oral glucose tolerance test (OGTT) and insulin tolerance test (1ST), body weight, abdominal fat, holding power, such as ultrasonic index to evaluate high fat and sugar to induce insulin resistance model after the build is successful, the intensity in the gym, aerobics, MSTN antibodies intervention,climb the ladder of strength,MSTN, strength and aerobic exercise combined with 5 kinds of sports in gym exercise intervention of model group rats, by blood flow dynamics,kinetics of ultrasonic, holding power and other indicators, biochemical indexes such as enzyme-linked immune response;morphological indexes such as HE staining,MASSON staining and immunohistochemical.Molecular techniques for testing such as Western Blot, RT-PCR and others.It is used of analysing that different groups within the rat cardiac changes and the key molecular protein gene expression of the insulin receptor, myocardial MSTN/PI3K7Akt in the signal path.Results:1. The resistance movement and the aerobic exercise and joint research and analysis on the improvement of the insulin resistance of myocardial cellsResistance exercise, aerobics, aerobics resistance movement joint can significantly reduce insulin resistance rats weight and heart weight (P<0.01), increased heart coefficient (P<0.05, P<0.05, P<0.01), decrease the Lee’s index (P<0.01). Ultrasonic detecting results and aerobic exercise combined aerobic and resistance exercise of left heart ejection fraction (EF), left heart shortening fraction (FS) and heart rate (HR) there was no significant influence; The resistance movement significantly improved EF (P<0.01), to the FS, the HR there was no significant change. The resistance movement and MSTN antibody interference and joint research and analysis on the improvement of the insulin resistance of myocardial cellsResistance movement and MSTN antibodies and their combined intervention scheme can significantly reduce insulin resistance in the rat body weight, heart weight (P<0.01, P<0.05), improve heart coefficient (P<0.05), reduce the body length (P<0.05); And the resistance movement and joint group can significantly reduce the Lee’s index (P<0.01). Aerobic and resistance exercise intervention of insulin resistance rats myocardial signal path are not completely consistent.Aerobic exercise is mainly improving insulin resistance rats cardiac symptoms by improving the IRS-1/PI3K/Akt signaling pathway,then resistance movement gets through MSTN and its receptor and PI3K/Akt, increasing the muscle area, improving insulin resistance. The two signaling pathways are in intersection, but not identical. The effects of two exercises are similar on cardiac function, increasing the myocardial area and the improvement of the inhibition of collagen fibrosis. Resistance exercise has obvious effect on protein expression and cell growth. Aerobic exercise mainly works through reducing the occurrence of sugar dysplasia and promoting the synthesis of glycogen. The effect that aerobic exercise loses weight is most obvious,the joint of aerobic exercise and resistance has no expression of superposition effect.2. The resistance movement and MSTN antibodies are paralleled to improve cardiac function, increase the myocardial area, inhibition of collagen fibrosis.They have same signal transduction mechanism and the joint of both intervention donnot have the stacking effect on myocardial insulin resistance. MSTN antibodies can improve myocardial insulin resistance.It infers that resistance improvs insulin resistance of cardiac function in rats through MSTN pathways.
