Objective:this study established an aging model by injecting D-galactose,then using SIRT1 agonist and inhibitor to explore the effects of the resistance exercise combined with SIRT1 on miRNAs and testosterone in aged mice.Methods:the objects of this experiment are males,SPF,3 months C57BL/6 mice,free feeding.Two weeks later,we injected 15%D-galactose(D-Gal)into the mice for 8 weeks to build aging model,the D-Gal solution were prepared with 0.9%saline,according to the dose of 500 mg/kg/D neck back subcutaneous injection.After modeling,aged mice were randomly divided into 6 groups:control(OC),resistance exercise(OR),EX627 injection(OE),resveratrol injection(OS),combination of resistance exercise and EX527 injection(OER),combination of resistance exercise and resveratrol injection(OSR).Fluorescence quantitative PCR was used to measure mir-1,-133a/b,-181,-206,-486 and-499 in mice,and ELISA was used to detect the contents of the mouse gastrocnenone.Results:(1)Multi-factor analysis of variance was used to analyze the data.It was found that resistance exercise factor has a significant effect on miR-1,-133a/b,-499(P<0.01),SIRT1 factor has a significant effect on miR-1,-133a/b,-486(P<0.01),and exercise combining SIRT1 interaction effect has a significant effect on miR-133b(P<0.05);compared OR group with OC group,the resistance exercise factor significantly reduced miR-1,-133a/b,-499(P<0.01),and the effect on miR-181,-206 and-486 was not significant(P>0.05);compared with OC group,It was found that in OE group and OS group,the SIRT1 factor significantly up-regulated miR-1 and-133a/b(P<0.01),and decreased miR-486(P<0.01),and the effect on miR-181,-206 and-499 was not significant(P>0.05);compared OER group and OSR group with the OC group,the exercise binding SIRT1 significantly reduced miR-133b(P<0.05),and the effect on miR-1,-133a,-181,-206,-486 and-499 was not significant.(2)It was found that the influence of exercise combined with SIRT1 interaction effects was significant on testosterone(P<0.05);compared with OC group,OSR group was significant difference in testosterone,testosterone rise significantly(P<0.05).Conclusion:1)miR-1,-133a/b and-499 were involved in the changes of skeletal muscle induced by resistance exercise,and the effect of resistance exercise on testosterone was not obvious.2)miR-1?-133a/b?-486 participated in the regulation pathway of SIRT1 in skeletal muscle of senile mice,while testosterone and SIRT1 interaction is not obvious.3)The adaptive change of the four-week resistance exercise combined with SIRT1 induced miR-133b and testosterone in aging skeletal muscle is more obvious than the two alone.
