Objective:Semiliquidambar cathayensis H.T Chang is a plant of Hamamelidaceae Semiliquidambar,Which only exists in China.This genus consists of three species and three variations.In order to study the Chemical Constituents from the leaves of Semiliquidambar cathayensis and the material basis of it,this paper reported systemically the isolation,identification and biological activities of S.cathayensis.Methods:Firstly,the paper tested tyrosinase inhibitory activities and the content of total phenol and total flavonoids of extracts from different leaf shape of S.cathayensis.Basing on the results of activities research,the compounds were isolated and purified by various chromatographic techniques and pre-HPLC.Their structures were identified on the basis of physicochemical characters and spectroscopic analysis.And the paper evaluate monomeric compounds 1-14,17-18,20 isolated for tyrosinase inhibitory activities.At last,through MTT test,the paper select extracts of different part of isolation for MH7A cell inhibitory activities.Results:Through basic activities research,among crude extracts,the full lotus leaf of S.cathayensis have most significantly tyrosinase inhibitory activity,with IC50value of 17.06 mg/ml,and the total phenol and total flavonoids were consistent with their tyrosinase inhibitory activity.Twenty compounds were isolated from the acetone extract of S.cathayensis leaves,which including 17 compounds of flavonoids,2 compounds of phenolic acids,1 compound of phenol glycoside.The compounds were identified as gallic acid?1?,quercetin?2?,gallic-catechin?3?,catechin?4?,kaempferol-3-O-?-L-rhamnoside?5?,kaempferol-3-O-?-L-arabinoside?6?,isoquercitrin-6''-gallate?7?,catechin-[8,7-e]-4?-?3,4-dihydroxy-phenyl?-dihydro-2?3H?-pyranone?8?,iso-myricitrin?9?,kaempferol-3-O-?2''-O-acetyl?-?-D-galactopyranoside?10?,kaempferol-3-O-?2''-O-acetyl?-?-D-glucoside?11?,kaempferol?12?.5-O-caffeoylquinic acid?13?,4-hydroxy-2-methoxyphenol-1-O-?-D-?6?-O-galloyl?-glucopyranosideand4-hydroxy-3-methoxyphenol-1-O-?-D-?6?-O-galloyl?-glucopyranoside?14?,Rutin?15?,quercitrin-3-O-?-D-galactopyranoside?16?,iso-quercitrin?17?,kaempferol-3-O-?-D-rutinoside?18?,kaempferol-3-O-?-D-glucopyranoside?19?,6??-Galloyl myricetin-3-O-?-D-glucopyranoside?20?.The results of tyrosinase inhibitory activity experiments showed that flavonoids?compounds2,3,5,12,20?have significantly activity,with IC50 value less than 0.96 mg/ml.Last MTT experiments showed that water phase of 40%methanol extraction have MH7A proliferation inhibition effect.Conclusions:All compounds are isolated from this plant for the first time.The 40%methanol extraction part have MH7A proliferation inhibition effect.The extraction of S.cathayensis leaves have significantly tyrosinase inhibition effect.It is especially important that the leaves of its non-medicinal parts can be developed into antioxidants and tyrosinase inhibitors.
