In order to find the optimum extraction and purification techniques, we study on the chemical constituents in the peels of Carya cathayensis Sarg. to provide a scientific basis for the further exploitation and utilization.An experiment was conducted to study on the chemical constituents in peels of Carya cathayensis Sarg. by three methods: the test tube method, PPC method and TLC method. And the chemical constituents such as amino acids, polysaccharides, flavonoids, saponin, volatile oil, cardiac glycoside, and alkaloids were identified.Effects of extraction temperature, ethanol concentration, ratio of solvent to sample (v/w), extraction hours and extraction times on the extraction yield of total flavonoids, polysaccharides, total alkaloids were analyzed by the orthogonal test. The results indicated that the optimum extraction conditions of total flavonoids were heating for 1hour at 60℃water bath for 3 times, ratio of solvent to sample 10:1 (v/w). And the maximum extraction yield is 1.09%. The optimum extraction conditions of polysaccharides were heating for 3 hours at 90℃water bath for 3 times, ratio of solvent to sample 10:1 (v/w). And the maximum extraction yield is 1.02%. The optimum extraction conditions of total alkaloids were heating for 1.5 hours at 60℃water bath for 2 times, ratio of solvent to sample 10:1 (v/w), ethanol concentration 50% .And the maximum extraction yield is 0.73%.Furthermore, the total flavonoids and total alkaloids were purified by macroporous resin NKA-9. The optimum purification conditions of total flavonoids were ethanol concentration 50%, sample quantity 2.08mg, flow velocity 6BV/h, sample pH 2. And the optimum purification conditions of total alkaloids were ethanol concentration 60%, sample concentration 2.67mg/ml, flow velocity 2BV/h, sample pH9.
