Juglans Cathayensis Dode. (Juglandaceae) grows in Shennongjia mountainous area of Hubei province of China. The leaves of the plant have been used as a folk medicine for the treatment of cancer. In our anti-tumor screening experiment, the n-BuOH-soluble fraction partitioned from the EtOH extract of Juglans Cathayensis Dode showed significant tumor inhibitory effects, which prompted us to initiate a chemical investigation of the n-BuOH-soluble fraction of this plant. As a result, 18 compounds have been isolated and 16 of them are identified on the basis of the physical constants and spectroscopic analysis. The structuresofthemareJucaletherA(1);(1R,2R,4R)1,2,3,4-Tetrahydronaphthalene-l,2,4-triol(2);3-Hydroxy-cyclobutanecarboxylicacid(3);4-oxo-5-hexenoicacid(4);(-)-regiolone(5);O-α-D-glucopyranosyl-[1,1]-α-D-glucopyranoside(6);α-L-rhamnose(7);5α,8α-epidioxy-6,22-diene-3β-ol(8);5α,8α-epidioxy-6,9(11),22-trien-3β-ol(9);kampferol(10);quercetin(11); ,7,8,4′-tetrahydroxyflavone(12);β-sitosterol(13);octacosylalcohol(14);14-hydroxy-octadecanoicacid(15) and 12-oxo-octadecanoic acid(16).Comp.1, 2 and 3 are novel compounds, the stereochemical structure of Comp.2 was established based on NMR spectra, FAB-MS, X-ray, et al. Comp.4, 6, 8, 9, 12, 15 and 16 were isolated from Juglandaceae for the first time. Comp.5, 7, 11 and 14 were isolated from this plant for the first time. The anti-tumor screening experiments reveal that Comp. 2,3,5 and 12 have significant cytotoxicities against the tumor cells. Which provide forceful support for the notion that these compounds should be further explored as a possible chemotherapeutic drug in terms of its possible effectiveness in treament of cancer.
