Objective:1.To observe the effect of electroacupuncture on motor function in MCAO rats.2.To verify the effect of electroacupuncture on spike activity of cerebral ischemic penumbra neurons in MCAO rats.Methods:Thirty-two male SD rats were divided into the normal group,the sham group,the model group and the electroacupuncture group.The animal model of MCAO was made by Zea Longa suture method The rats in the electroacupuncture group were intervened by electroacupuncture at Fengchi acupoint on both sides.Bederson modified neurological severity score was used to assess neurological deficits.CatWalk gait analysis system was applied to evaluate limb movement function before and after modeling and before and after the treatment.Cerebus system was used to detect spike frequency of neurons.6.It was verified by TTC staining.Result5:1.Weight:The weight of the non-MC AO model group increased,and the non-MCAO model group had weight loss.The speed of weight loss of electroacupuncture group was modified after 3 days treatment.2.Bederson modified neurological severity scores showed that the neurological deficit signs appeared in the MCAO model rats after modeling.and there was a statistically significant difference between the non-MCAO model group and the MCAO model groupp(P<0.05).After 24 hours of modeling and 3 days later,the Bederson modified neurological severity scores in the MCAO model group showed a downward trend,but there was still a certain degree of neurological deficit,which had a statistically significant difference with the non-MCAO model group(P<0.05)There was a statistically significant difference between the model group and the electroacupuncture group(P<0.05).3.CatWalk gait results:? Average movement speed and duration:The movement speed of MCAO model rats was significantly slower than that of non-MCAO model rats(P<0.05),but the duration of MCAO model rats increa sed,and the non-MCAD model did not.After 3 days,the movement speed of electroacupuncture group was faster and the duration vas shorter than those of the model group.There were statistically significant differences between these two groups(P<0.05).? Maximum Contact Area(MCA):24 hours after modeling and 3 days later,there was no statistically significant difference between MCAO model rats and non-MCAO model rats in MCA of both fore limbs(P>0.05).24h after modeling,MCAO model rats showed the statistical differences in MCA of both hind limbs compared with non-MCAO model rats(P<0.05).After 3 days MCAO model rats showed no statistical difference in MCA of both hind limbs compared with non-MCAO model rats(P>0.05).?Paw Print Area-(PA):24h after modeling and 3d later,there was no statistical difference between MCAO model rats and non-MCAO model rats in PA of both fore limbs(P>0.05),24 hours after modeling,there were statistically significant differences between MCAO model rats and non-MCAO model rats in PA of both hind limbs(P<0.05).After 3 days,there was no statistically significant difference between MCAO model rats and non-MCAO model rats in PA of both hind limbs(P>0.05).?MCA,PA ratio:The ratio were ralated to MCA and PA,24h after modeling:MCAO model rats in MCA,PA of both fore limbs showed a downward trend.After 3 days:there was no significant difference in ratio of the fore limbs between groups.?Mean Intensity of Pressure(MI):After MCAO modeling:there were statistically significant differences in MI of both fore limbs between MCAO model rats and non-MCAO model rus(P<0.05).After 3 days:there was no significant difference in MI of the right fore limb between MCAO model rats and non-MCAO model rats(P>0.05).The MI of the left fore limb in the model group was significantly different from that in the normal group,the sham operation group and the electroacupuncture treatment group(P<0.05),24 hours after MCAO modeling and 3 days later,there was no significant difference in MI of the right hind limb between MCAO model rats and non-MCAO model rats(P>0.05).After MCAO modeling,there were statistically significant differences between the MCAO model rats and the non-MCAO model rats in MI of the left hind limb(P<0.05).After 3 days:there was no significant difference in MI of the right hind limb between MCAO model rats and non-MCAO model rats(P>0.05).? Single foot standing time:After MCAO modeling:there were statistically significant differences between the MCAO model rats and the non-MCAO model rats in the single foot standfing time of both fore limbs and hind limbs(P<0.05).After 3 days,the single foot standing time of bath fore lim bs and hind limbs in the model group was significantly difffferent from those in the other three groups(P<0.05)4.Results of spike Telease in M1 area:the spike ielease was rhythmic "before modeling.After MCAO modeling,spikes' original rhythm changed and distributed diffusely.After 3 days,spike frequency increased.5.TTC staining:Normal brain tissue were stained dark red.Rats in the model group had obvious white.ischemic focuses,which were stained white.Conclusious:1.The electroacupuncture treatment has a positive effect on imeproving the Bederson modified neurological severity scores,and it is consistent with the overall results of Catwalk gait analysis,such as MCAA/PA ratio of hind limbs,sppeds,durations and stands.2.The MCA/PA ratio of all lim bs.expecially the cross limbs changed obviouslly after modeling,and recovered with the improvement of coordination functions.MCA/PA ratio can reflect therecovery of limbs after stroke to a certain extent3.The electroacupuncture treatment can promote the recovery of Bedeuson modified neurological severity score in MCAO rats.It has significant effects on the recovery of motor function of limbs,especially in speed,duration and single foot standing time.4.The electroacupuncture treatment can improve spike frequency and spike patterns in M1 area of MGAO rats,improve motor function of rats after stroke.
