| Objective:To elucidate the mechanism of electroacupuncture in treating brain ischemic disease, it is studied that the influence of acupuncture at Quchi on mocrocirculatory blood flow of hippocampus and transport function of microvascular endothelial in MCAO rats.Methods:Brain ischemia was induced by middle cerebral artery occlusion with thread ligature in wistar rats(MCAO). These rats were randomly divided into four groups:sham operation groups , model groups, Diji groups and Quchi groups. The change of microcirculatory blood flow in hippocampal CA1 area was monitored by laser-Doppler Flow meter(LFD) on the first,fifth,tenth day after MCAO respectively in each group. Morphological changes of hippocampal neurons was observed in HE staining. Quantitative analysis of apoptoic cells in hippocampal CA1 area was carried out through image processing tool-Pro Plus version 4.5. The content of glucose,lactic acid,pyruvic acid in brain tissue was analyzed using measureing kit respectively. The expression of GLUT-1,MDR-1,HIF-1 was observed with the method of immunohistochemisty assay.Results: Microcirculatory blood flow in hippocampal CA1 area of MCAO rats was obviously increased after electroacupuncture at Quchi acupoint ,this effects were obveriously than the effects of Diji group. Apoptosis rate of neurons in hippocampal CA1 area MCAO rats in Quchi groups was decreased obviously than that in other groups. The content of lactic acid ,calcium and glutamate in brain tissue were decreased after acupuncturing at Quchi. The content of pyruvic acid and glucose was increased in Quchi groups. Expressing quantity of HIF-1αin hippocampal CA1 area of rats was increased after electroacupuncture. The effect of electroacupuncture on expressing of GLUT-1, MDR-1 was not obvious.Conclusions: Microcirculatory blood flow in hippocampal CA1 area of MCAO rats could be increased by acupuncture. Apoptosis rate of neurons in hippocampal CA1 area could be decreased through acupuncture and the apoptosis could be inhabited. Expression of HIF protein in hippocampal CA1 area of MCAO rats could be increased by acupuncture. Transporting function of microvascular endothelial cells could be improved and neuronal injury was relieved after cerebral ischemia by means of acupuncture. |
