| Objective:Based on Treg/Th17,the effect mechanism of Banxia Xiexin decoction on the eradication of Hp and the regulation of gastric mucosal immune microenvironment in hp-infected PLGC mice was investigated,providing a theoretical basis for the reversal of PLGC in traditional Chinese medicine.Research methods:Selection successful PLGC Hp infection mouse model and normal group 5 only 25,respectively,the normal group,the model group,the quadruplre therapy group,the whole party group,the Shengqing group,the Jiangzhuo group,5 only.Medication intervention: the normal group and model group were given normal saline of 10ml/kg/d for 6 weeks,followed by free diet and drinking water.The quadruplre therapy group was given quadruplre therapy 10ml/kg/d by gavage for 2 weeks,followed by free diet and drinking water.The whole party group,the Shengqing group and the Jiangzhuo group were given Banxia xiexin Decoction,the Shengqing Decoction and the Jiangzhuo Decoction 10ml/kg/d by gavage for 6 weeks respectively.Detection: put to death in mice after drug intervention,take blood,full stomach,Hp by HE staining and Giemsa staining counting,observed the pathological and histological changes of gastric mucosa by double antibody sandwich enzyme immunoassay(Elisa)test in mice blood and gastric tissue and serum cytokines in the TGF-?,IL-6,IL-17 A content,through the protein imprinting method(Western Blot)detection Fox P3,ROR?t protein expression in gastric tissue.Research results:1.In the aspect of Hp eradication: compared with model group than quadruplre therapy group effectively the effect of Hp eradication(P<0.05).The whole party group,the Shengqing group and the Jiangzhuo group compared with model group,no significant difference(P>0.05),between the whole party group,the Shengqing group and the Jiangzhuo group no statistical difference(P>0.05),but which has a tendency to the eradication of Hp in numerical,and the whole party group is superior to the Shengqing group and the Jiangzhuo group.2.In terms of improving gastric mucosa pathology: Banxia xiexin Decoction has the best effect.Compared with the model group,the whole party group and the Shengqing group effectively improved the chronic inflammatory response of gastric mucosa(P<0.05),while the whole party group and the Shengqing group showed no statistical difference(P>0.05).the chronic inflammatory response of gastric mucosa was significantly improved in the whole party group and the Shengqing group(P<0.01),and there was no statistical difference between the whole party group and the Shengqing group(P>0.05).There was no statistical difference between the model group,the quadruplre therapy grou and the Jiang Zhuo group(P>0.05),and the quadruplre therapy grou and the Jiang Zhuo group also showed a tendency to improve the chronic inflammatory response.Compared with the model group,gastric mucosal inflammation activity was significantly improved in all four groups(P<0.01),and there was no difference between all four groups(P>0.05).In terms of value,the improvement degree of Banxia xiexin Decoction and its prescription was more obvious.Compared with the model group,there was no statistically significant difference in improving gastric mucosa atrophy and PLGC between all four groups(P>0.05),and there was no statistically significant difference between all four groups(P>0.05).Both groups showed a numerical trend of improvement,and Banxia xiexin Decoction had the best overall efficacy.3.In the molecular mechanism of action,the quadruplre therapy group,the whole party group,the Shengqing group and the Jiangzhuo group,all four groups can decrease the TGF-?,Fox P3,IL-6,ROR?t,IL-17A(P<0.05).There is a trend that the whole team is superior to the demolition team.Immunosuppressive Treg:TGF-? serum: Compared with the model group,TGF-? in serum was effectively reduced in all four groups(P<0.01),and there was no statistical difference between the four groups(P>0.05),but in the numerical trend,the quadruplre therapy group was the best,followed by the Shengqing group,the whole party group and the Jiangzhuo group.TGF-? in gastric tissue homogenate: compared with the Shengqing group,TGF-? in gastric tissue homogenate was significantly reduced in the quadruplre therapy group(P<0.01),and compared with the whole party group,the effect was the best in the quadruplre therapy group(P<0.05).There was no statistical difference between the whole party group,Shengqing group and Jiangzhuo group(P>0.05).In numerical comparison,the Jiangzhuo group showed a better trend than the whole party group and the Shengqing group,followed by the whole party group and the Shengqing group.Fox P3 expression level: Compared with the model group,Fox P3 expression level was significantly decreased in the quadruplre therapy group,the whole party group and the Shengqing group(P<0.01),while Fox P3 expression level was significantly decreased in the Jiangzhuo group(P<0.05).Compared with the Jiangzhuo group,Fox P3 protein expression level was significantly decreased in the quadruplre therapy group(P<0.05).There was no statistical difference between the whole party group,the Shengqing group and the the Jiangzhuo group(P>0.05).In numerical comparison,the quadruplre therapy group showed the best reduction in Fox P3 expression,followed by the whole party group,the Shengqing group and the Jiangzhuo group.In the immune reaction Th17: serum IL-6 concentration: Compared with the model group,serum IL-6 concentration in all four groups was significantly reduced(P<0.01).Compared with the Shengqing group,the serum IL-6 concentration in the quadruplre therapy group was significantly reduced(P<0.01).Compared with the whole party group and the Jiangzhuo group,the serum IL-6 concentration in the quadruplre therapy group was effectively reduced(P<0.05).There was no statistical difference among the whole party group,the Shengqing group and the Jiangzhuo group(P>0.05).In numerical comparison,the Jiangzhuo grou showed a better trend than the whole party group and the Shengqing group,followed by the whole party group and the Shengqing group.Il-6 concentration in gastric tissue serum: Compared with the model group,il-6 concentration in gastric tissue serum was significantly reduced in all four groups(P<0.01).Compared with the Shengqing group and the Jiangzhuo group,the quadruplre therapy group effectively reduced the concentration of IL-6 in gastric tissue serous fluid(P<0.05).There was no statistical difference between the whole party group,the Shengqing group and the Jiangzhuo group(P>0.05).In numerical comparison,the whole party group showed a trend of being superior to the Shengqing group and Jiangzhuo group.The expression level of ROR?t protein in the quadruplre therapy group and the whole party group was significantly lower than that in the model group(P<0.01),while the expression level of ROR?t protein in the Shengqing group and the Jiangzhuo group was significantly lower(P<0.05).There was no statistical difference between the four groups(P>0.05).In numerical comparison,the whole party group was superior to the other three groups,followed by the quadruplre therapy grou,the Shengqing group and the Jiangzhuo group.Serum Il-17 A concentration: compared with the model group,the serum Il-17 A concentration in all four groups was significantly reduced(P<0.01),and there was no statistical difference between the four groups(P>0.05).In numerical comparison,the Jiangzhuo group was superior to the other three groups,followed the quadruplre therapy group,the whole party group and the Shengqing group.Il-17 A concentration in gastric tissue homogenate: Compared with the model group,Il-17 A concentration in gastric tissue homogenate was significantly reduced in the quadruplre therapy group,the whole party group and the Shengqing group(P<0.01),while Il-17 A concentration in gastric tissue homogenate was effectively reduced in the Jiangzhuo group(P<0.05).The concentration of IL-17 A in the gastric tissue homogenate was significantly reduced in the quadruplre therapy group compared with that in the Shengqing group and the Jiangzhuo group(P<0.01),and the concentration of IL-17 A in the gastric tissue homogenate was effectively reduced in the quadruplre therapy group compared with that in the whole party group(P<0.05).There was no statistical difference between the whole party group,the Shengqing group and the Jiangzhuo group(P>0.05).In numerical comparison,the whole party group showed a trend of being superior to the Shengqing group and the Jiangzhuo group.Conclusion:Banxia Xiexin decoction can effectively eradicate Hp infection,improve gastric mucosa histopathology,regulate gastric mucosal immune balance,and reduce inflammatory response by lifting and lowering turbidity,which has certain significance for delaying PLGC process in the pathogenic pathway of "Bacteria-Inflammation-Cancer". |
