Objective:To observe the effects of Xiao Ye Zeng Sheng Fang on the morphology,pathology and estrous hormones and their receptors in the mammary gland of rats with mammary gland hyperplasia.Method:Sixty SD rats were randomly divided into blank control group,model group and treatment group.On the first day of adaptive feeding in each group of rats,the estrus smear was observed at 9am and 16pm daily by the observation of the estrus smear at the 5th and 10th day of the treatment,and the estrous interval was observed.Blood was taken from the internal iliac vein and E2,P and PRL were detected.In the modeling stage,the blank control group was intramuscularly injected into the lateral hind limb of rats.The model group and the treatment group were intramuscularly injected into the lateral hind limbs of rat mammary gland hyperplasia model by intramuscular injection of estradiol benzoate.In the treatment stage,the blank control group and the model group were applied with the starch dextrin on the third pairs of breasts of the rats,and the treatment group was applied to the third pairs of breasts of the lobular hyperplasia.On the fifth day of treatment,the third pair of right breasts of each group of rats was taken.On the tenth day of treatment,the third pair of left breasts of each group of rats were taken,and the specimens were subjected to paraffin section and HE staining for pathological diagnosis.After the treatment,the diameter of the nipple,the area of breast acinus and the diameter of the catheter were observed.The pathological changes of the mammary gland in each group were observed by HE staining.The levels of sex hormones?E2,P and PRL?were detected by enzyme-linked immunosorbent assay.Detection of rat sex hormone receptor?ER,PR?by immunohistochemistry.Results:?1?Diameter of nipple:Before the model was established,there was no significant difference in the diameter of the nipple between the three groups ?P=0.2053?.After modeling,the nipple diameter of the model group and the treatment group was increased compared with the control group?P<0.0001;P=0.0013?.After 5days of treatment,the nipple of the model group was still increased compared with the control group?P<0.0001?,but the diameter of the nipple of the treatment group was smaller than that of the model group?P=0.0004?.After 10 days of treatment, compared with the control group,the nipple of the model group still increased ?P=0.0008?,but the diameter of the nipple of the treatment group was further reduced?P=0.0002?,and returned to the nipple level of Normally,the ductal epithelial cells are arranged neatly;the glandular lobes and the small and medium-sized ducts are surrounded by fibroblasts,and the interlopes are separated by adipose tissue,and the cell morphology and arrangement are normal.In the model group,there were enlarged glandular epithelial hyperplasia and secretions in the duct,and a few acinar were slightly dilated and showed moderate hyperplasia.In the treatment group,the individual acinar showed mild hyperplasia,no dilatation,and secretions in the acinus and duct.After 10 days of treatment:the control group saw the mammary gland at rest,the number of glandular leaves was small,the acinar cells in the glandular lobes were tight,the glandular epithelial cells were arranged neatly,the myoepithelial cells were intact,the mammary gland lobes were scattered,and the acinar cavity did not expand.In the model group,the lobular hyperplasia of the breast was enlarged,the volume was still enlarged,the number of acinar cells was increased,the glandular epithelial hyperplasia increased the acinus and the secretion in the duct,and some acinar and duct were in an expanded state.In the treatment group,there was no obvious hyperplasia of the lobule of the breast,and some acinar had mild hyperplasia and no expansion.?10?Breast acinar area:After 5 days of treatment,the mammary gland area of the model group and the treatment group increased compared with the control group ?P<0.0001,P<0.0001?,but the acinar area of the treatment group.Less than the model group?P=0.0038?;after 10 days of treatment,compared model of hyperplasia of mammary glands was successfully replicated by intramuscular injection of estradiol sequential progesterone.The ammary gland hyperplasia rats showed an increase in the diameter of the nipple.Under the microscope,the area of the breast acinus and the diameter of the catheter increased,E2 level and PRL level increased,the ER expression was more significant,the estrous cycle duration was prolonged,and some rats'estrus were disappeared. fter 10 days of withdrawal of exogenous sex hormones,the breast model has not recovered.Exogenous sex hormones had no significant effect on the expression of P and PR in rats.The occurrence of mammary gland hyperplasia is related to the isorder of hormone secretion in the body,especially related to the elevated levels of E2 and PRL.The external breast lobular hyperplasia screening can accelerate the ejuvenation of the mammary gland hyperplasia rats,which shows that the diameter of the nipple is reduced,and the area of the mammary gland is reduced under the microscope.There is no significant effect on the inner diameter of the catheter,and the prolonged estrous cycle is shortened.Xiao Ye Zeng Sheng Fang can reduce the unnormal E2?PRL levels,reduce the stimulation of estrogen on the mammary gland,and provide experimental evidence for the clinical application of drugs.
