Effect Of Danhong Acupoint Injection On NO In Serum And Expression Of VEGF/KDR In Uterine Tissue Of Female Rats With Qi Stagnation And Blood Stasis Syndrome

Objective:To explore the effect of Danhong Acupoint Injection on qi stagnation and blood stasis syndrome and its possible mechanism.Methods:Forty healthy SD female rats in 6-8 weeks were randomly divided into four groups:blank group,model group,traditional Chinese medicine group and acupoint injection group,with 10 rats in each group.Except for the blank group,the other three groups of rats were injected with adrenaline hydrochloride(ADR)in the left lower abdomen for 18 consecutive days and received unforeseen stimulation for 4hours to reproduce the model of qi stagnation and blood stasis in female rats.24 hours after the completion of the model,the Chinese medicine group was infused with Taohong Siwu Decoction once a day for 14 times;the acupoint injection group was injected with Danhong injection once every 3 days for 4 times.After 2 weeks of treatment,the uterus of rats was extracted after the blood was collected from the heart.The changes of hemorheology,blood coagulation and serum NO were observed.The expression of VEGF/KDR protein and RNA in rat uterus was detected by immunohistochemistry and real-time fluorescence quantitative PCR.Results:Compared with the blank group,the whole blood(high,medium and low)viscosity,plasma viscosity and hematocrit of Hemorheology in the model group were significantly increased(P<0.01 or P<0.001),thrombin time(TT)and activated partial thromboplastin time(APTT)were significantly shortened,fibrinogen(FIB)content was significantly increased(P<0.01 or P<0.001),there was no significant difference in Prothrombin time(PT)(P>0.05),and serum NO expression was significantly decreased(P<0.01),and the expression of VEGF and KDR protein andm RNA in uterine tissue decreased significantly(P<0.05).Compared with the model group,the hemorheology whole blood(high,medium,low)viscosity,plasma viscosity,hematocrit of female rats in the traditional Chinese medicine group and acupoint injection group decreased significantly(P<0.001 or P<0.05),TT,APTT prolonged significantly,FIB content decreased significantly(P<0.05 or P< 0.01),there was no significant difference in PT(P>0.05),serum NO content increased significantly(P<0.01),and the expressions of VEGF and KDR protein and m RNA in uterine tissue increased significantly(P<0.05),and there was no statistical difference between the two groups.Conclusion:(1)Danhong injection was used to treat qi stagnation and blood stasis syndrome in female rats by acupoint injection,and the effect of activating blood circulation and removing blood stasis was achieved by reducing blood viscosity and improving coagulation function;(2)The effect of removing blood stasis and generating new blood stasis was achieved by promoting the increase of no content in serum and the expression of VEGF and KDR in uterine tissue;(3)the mechanism of action might be that serum NO,VEGF and KDR were improved in uterine group The important factors of weaving blood flow state promote each other,so as to achieve the functions of "activating blood circulation and removing blood stasis" and "removing blood stasis and generating new".
