This paper included two parts:the chemical composition and quality standard of Xianzhuli.Xianzhuli is a light yellow to reddish brown liquid juice extracted from the Gramineae plant bamboo after processing.It has bamboo aroma,slightly sweet taste,and has the functions of clearing heat and reducing phlegm and relieving horror.It can also be called bamboo oil,bamboo vinegar and bamboo juice.Currently,it is mainly used in clinical treatment of phlegm fever cough,hiccup,asthma and bronchitis.Studies have shown that the chemical composition of Xianzhuli mainly contains flavonoids,amino acids,inorganic elements,phenolic acids and other components.In this thesis,We continued to study the chemical composition of the n-butanol portion based on the previous research on the chemical composition of Xianzhuli,Using thin-layer chromatography,C18 column chromatography,HPLC technology,and related natural product isolation and purification methods,19compounds were isolated and identified from the fresh bamboo n-butanol extraction site,respectively:syringaresinol 4?-O-?-D-glucopyroside(1),5,5'-dimethoxylaricires inol-4'-O-?-D-glucopyranoside(2),(+)-lyoniresinol-3?-O-?-D-glucopyranoside(3),(-)-4-epi-lyoniresinol(4),(-)-lyoniresinol-2?-O-?-D-glucopyranoside(5),(+)-lyoniresinol(6),8,8'-bisdihydrosiringenin glucoside(7),2,4,6-trimethoxyphenyl1-O-?-D-glucopyranoside(8),threo-8S-7-meth-oxysyringylglycerol(9),salidroside(10),(7S,8R)syringoylglycerol(11),syringaldehyde(12),2,6-dimethoxy-1,4-benzoquinone(13),p-Hydroxybenzaldehyde(14),n-butyl-?-D-fructopyranoside(15),n-butylpyroglutamate(16),asterbatanoside A(17),(+)-ovafolinin B-9?O-?-D-glucopyranoside(18),Propiophenone4?-O-Primeveroside(19).Compounds 1~5,7~11,15~19 were isolated from Xianzhuli for the first time.As a traditional Chinese medicinal material,Xianzhuli has a low level of current standard quality control,which is difficult to effectively control its quality.In the existing reports,there are few studies on the effective substances in Xianzhuli,and it is difficult to determine the quantitative active index components in the existing studies.In the early extraction and separation of the chemical components of Xianzhuli,we found that lignans were the main chemical components.Among them,(+)-lyoniresinol-3?-O-?-D-glucopyranoside is the main characteristic component isolated from Xianzhuli.In this paper,thin-layer chromatography was used to qualitatively identify(+)-lyoniresinol-3?-O-?-D-glucopyranoside in Xianzhuli and a method for the determination of(+)-lyoniresinol-3?-O-?-D-glucopyranoside in Xianzhuli by HPLC was established,which can optimize the quality standard of Xianzhuli.The chromatographic conditions were:Hypersil C18 column(5?m,250mm×4.6mm);mobile phase was acetonitrile-water(12:88);flow rate was 1.0 ml/min;detection wavelength was 220 nm;column temperature was 30°C;injection volume is 10?l.The results showed that the injection amount of(+)-lyoniresinol-3?-O-?-D-glucopyranoside was in the range of 0.0435~0.6954?g,and the peak area showed a great linear relationship with the injection amount.Regression equation:Y=2700.2983X+14.3918,R~2=0.9999(n=6).The method has been verified by methodological verifications such as stability,precision,reproducibility and recovery rate.which shows that this method is accurate,reproducible,simpleand.so it can be used as quantitative detection method of(+)-lyoniresinol-3?-O-?-D-glucopyranoside.In addition,the total solids,relative density,and pH determination in 36 batches of Xianzhuli were studied in this thesis,and reasonable inspection limits were established.
