Objective: To study the effects of different intervention methods(acupuncture,scraping)on CK and LDH in serum,morphology change of skeletal muscle and expression of desmin of the rats with chronic skeletal muscle injury model,so as to explore the mechanism of improvement and its differences of different intervention methods on chronic skeletal muscle injury.Methods: Forty healthy SD rats with chronic skeletal muscle injury model using the classical rat downhill eccentric contraction exercise model were selected and randomly divided into a model control group,a natural recovery group,a scraping group,and an acupuncture group,with 10 rats in each group.And the acupuncture group was treated with acupuncture therapy,the scraping group was treated with local scraping,treated for 7 days and taken the using part.Meanwhile,the model control group was not treated,executed after the model was completed;the natural recovery group was not treated,too.The rats of the group moved free for 7 days in the cage after the model was completed,and then taken the using parts of its;As for the blank control group,it didn't run and kept moving free in the cage until the using part was taken.At the same time,enzyme-linked immunoassay(ELISA)was used to detect the changes of the activity of CK and LDH in serum of the rats with chronic skeletal muscle injury model;hematoxylin-eosin staining(HE)method was used to observe the morphology change of the lateral femoral muscle of the rats with chronic skeletal muscle injury model;immunohistochemical techniques were used to qualitatively analyze the expression of desmin in the lateral femoral muscle of rats with chronic skeletal muscle injury model in each group;western Blot method was used to quantitatively analyze the expression of desmin in the lateral femoral muscle of rats with chronic skeletal muscle injury model in each group.Results:1.ELISA was used to detect the variational situation of the activity of CK and LDH in serum of the rats with chronic skeletal muscle injury model by different intervention methods C K:(1)As the treatment progressed,its activity in serum of the rats belong to the acupuncture group and the scraping group decreased significantly,and both are significantly lower than the model control group(P <0.05),compared with the natural recovery group,the difference is also significant(P <0.05),indicating that the therapeutic methods of acupuncture and scraping both can obviously reduce the activity of CK in serum of the rats with chronic skeletal muscle injury model,and its effect is significantly better than that of the natural recovery group.(2)Compared with the scraping group,the acupuncture group has a significant difference,which has a statistical sense(P <0.05),showing that the effect of acupuncture towards the activity of CK in serum of the rats with chronic skeletal muscle injury model is obviously better than that of the therapeutic methods of scraping.LDH:(1)The activity of LDH in serum of the rats in the acupuncture group and the scraping group significantly decreased with the progress of the treatment,which is significantly different from the model control group(P <0.05),compared with the natural recovery group,the difference has a statistical sense(P <0.05),indicating that acupuncture and scraping therapy both can significantly reduce the activity of LDH in serum of the rats with chronic skeletal muscle injury model and its effect is significantly better than that of the natural recovery group.(2)Compared with the scraping group,the acupuncture group has a significant difference(P <0.05),indicating that the effect of acupuncture towards the activity of LDH in serum of the rats with chronic skeletal muscle injury model is significantly better than that of scraping treatments.2.Hematoxylin-eosin staining(HE)method was used to observe the effect of different intervention methods on the morphology change of lateral femoral muscle tissue of the rats with chronic skeletal muscle injury model Microscopic observation of lateral femoral muscle tissue microtome sections of SD rats: The muscle fiber of the blank control group array neat,all of its have consistent size,complete cell membrane,clear boundaries,uniform cytoplasm,and normal nucleus,without hyperplasia,hypertrophy or shrinkage,tissue edema and infiltration of inflammatory cells,horizontal stripes can be seen in the microtome sections;As for the model control group,muscle fibers are ruptured,with obvious necrosis and degeneration,disordered arrangement,different sizes,unclear cell boundaries,enlarged nucleus,and tissue edema,infiltration of inflammatory cells and fibrous tissue proliferation are obvious,as well as the expansion and congestion,and blurred horizontal lines in the microtome sections;For the natural recovery group,the muscle fibers are broken,and arrange disorderly,with different sizes and unclear cell boundary,the nucleus is slightly larger,the tissue edema is reduced,and the inflammation cell infiltration is slightly reduced,but the fibrous tissue proliferation is obvious;To the scraping group,the muscle cells are distributed evenly,the cell boundary is clear,the cytoplasm is uniform,and the nucleus is slightly larger,without tissue edema,the inflammatory cell infiltration is reduced,the fibrous tissue proliferation is improved,and there is no congestion;About the acupuncture group,there is no obvious degeneration of muscle fibers,with neat arrangement,clear cell boundaries,uniform cytoplasm,slightly large nuclei of a small number,slightly less infiltration of inflammatory cells,and mild proliferation.3.Immunohistochemical techniques were used to qualitatively analyze the expression of desmin in lateral femoral muscle of the rats with chronic skeletal muscle injury model under different intervention methods.Compared with the model control group,the difference of the expression of desmin in lateral femoral muscle of the rats in the acupuncture group and the scraping group is significantly,which both is higher than the model control group(P <0.05),compared with the natural recovery group,the difference was also significant(P <0.05),indicating that acupuncture and scraping treatment methods both can significantly increase the expression of desmin in lateral femoral muscle of the rats with chronic skeletal muscle injury model,and its effect is significantly better than that of the natural recovery group.And the difference between the acupuncture group and the scraping group is significant(P <0.05),showing that the effect of acupuncture towards the effects of the expression of desmin in lateral femoral muscle of the rats with chronic skeletal muscle injury model is significantly better than that of the scraping treatments.4.Western blot method was used to quantitatively analyze the expression of desmin in lateral femoral muscle of the rats with chronic skeletal muscle injury model under different intervention methods.The expression level of desmin in lateral femoral muscle of the rats in the acupuncture group and the scraping group both are higher than the model control group(P <0.05),compared with the natural recovery group,the difference is significant(P <0.05),indicating that acupuncture and scraping treatments can significantly improve the expression level of desmin in lateral femoral muscle of the rats with chronic skeletal muscle injury model,and its effect is significantly better than that of the natural recovery group.And the difference between the acupuncture group and the scraping group is significant(P <0.05),indicating that the effect of acupuncture towards the effects of the expression of desmin in lateral femoral muscle of the rats with chronic skeletal muscle injury model is significantly better than scraping treatments.Conclusions:1.Acupuncture and scraping both can reduce the activity of CK and LDH in serum of the rats with chronic skeletal muscle injury model,have a obviously improvement role on skeletal muscle tissue morphology and increase the expression of desmin in lateral femoral muscle.2.In contrast,the effect of acupuncture on reducing the activity of CK and LDH in serum of the rats with chronic skeletal muscle injury model,improving the morphology of skeletal muscle tissues and increasing the expression of desmin in lateral femoral muscle is significantly superior than scraping therapy.
