Skeletal muscle injury is a name of disease in western medicine, which belongs to the injury of the muscle and tender in traditional Chinese medicine.In western medicine, the injury of the muscle and tender is considered as soft tissue injury. The soft tissues includes muscle, tendon, ligament, subcutaneous tissue, muscle, joint capsule, the articular cartilage, intervertebral disc tissue and so on. Skeletal muscle injury is the most common injury in all the soft tissue injuries. According to a large number of clinical researches, Chinese medicine has good therapeutic effect on skeletal muscle injury, especially acupuncturing Ashi points. The treating course is short and the therapeutic effect is very satisfactory. It has become an effective method in treatments of skeletal muscle injury in traditional chinese medicine.Objects:The rats’weight changes, independent activity frequency and behavioral indicators were observed, after quickly stimulating the local injury points. At end of the treatment, deltoid, soleus muscle and musculi quadriceps femoris were observed by HE staining. Observing the differences of the effects of acupuncture at different time, according to the injury of skeletal muscle fibers in rats damage and abnormal staining cell aggregation. Observing the loss of desmin in skeleton proteins by immunohistochemistry, further confirmed the correlation between muscle repair and cytoskeletal protein, which indicated that acupuncture were positive induction of skeleton muscle tissue reorganization, and the muscle repair processes occur in advance.Methods:54male Sprague-Dawley rats were divided into3groups randomly:normal group(6rats), model group(24rats), treatment group(24rats). All the animals in model and treatment groups were given28days downhill running, slope was30degrees. Then divided the two groups into immediate model group,12h model group,24h model group,72h model group, immediate treatment group,12h treatment group,24h treatment group,72h treatment group, with6rats in each group. Observed and recorded the rats weight and independent activity frequency. According to the treatment named group,respectively in the model immediately,12h,24h,72h in rats on the right side of the deltoid muscle, soleus muscle, musculi quadriceps femoris were oblique inserted and lift-thrust3times.After that, deltoid, soleus muscle, and musculi quadriceps femoris were observed by HE staining and immunohistochemistry.Packet after drawing materials,the teeatment group the right muscle right treatment group, the control group was left, and named according to the time window, the same with the other groups were taken before.Results:1Eccentric exercise induced skeletal muscle injury in rats28days after modeling, significant difference of rats’weight existed between model group and normal group (P<0.01). There was obvious difference in the independent activity frequency of rats before and after modeling (P<0.05). Change trend of the experimental data reflected the relevant behavior of rats in each treatment group, and clearly reflected that the rats after modeling showed slowing growth in weight and decreasing of independent activities.2Immediately after the modeling, compared right side with left side in12h,24h,72h model groups after HE staining. Results showed that in all left parts inflammatory cells increased significantly, muscle fiber damaged, arrangement disordered. compared with the normal group, there was remarkable difference on right side, but better than left side. Between right side and left side, there was statistically significant difference in muscle fiber damage and abnormal staining cell aggregation (P<0.05).3According to the number of damaged muscle fiber and aggregation of abnormal staining cells at different time, deltoid and soleus muscle damaged less as time progresses on right side, however, on left side soleus and musculi quadriceps femoris damaged more as time goes on.4By immunohistochemistry, integrated optical density(IOD) of desmin in the normal group was higher than that of other groups, and most of them had significant difference (P<0.05). Comparison between the groups, IOD of the12h model group was lowest. Compared the groups of the same time point, IOD of right side in treatment groups was higher than left side and model group. Compared with each group of different time points in the same group,12h group showed relatively lower IOD.Conclusions:1This experiment confirmed the effect of oblique insertion of Ashi points on reparing skeletal muscle injury2Compared the acupuncture time, it was obseved that the immediate effect of24h and72h acupuncture were better.12h and instant therapy had poor effect.3The affected part of the same part of acupuncture side cannot be in short-term inside the muscle injury has obvious therapeutic effect.
